阿司匹林对人结肠癌HT-29细胞肠分化标志物表达的影响
[Abstract]:Background & AIM: nonsteroidal anti-inflammatory drug aspirin is effective in the prevention and treatment of colon cancer. In recent years, it has been considered that the prevention and treatment of colon cancer can be achieved through the regulation of colon cancer stem cells. When colon cancer stem cells mature, colon cancer will no longer progress, or even fade. The aim of this study was to investigate the effect of aspirin, a non-steroidal anti-inflammatory drug, on the differentiation of human colon cancer cell line HT-29. Methods: the effects of aspirin at different concentrations on the proliferation of HT-29 cells were detected by living cell count kit (CCK-8). The expression of m RNA in intestinal differentiation marker mucin 2 (mucin 2 MUC2), trefoil factor 3 (trefoil factor 3tff3, sucrose enzyme isomaltase (sucroseisomaltase) and lysozyme (lysozyme) were detected by real time real-time fluorescent quantitative polymerase chain reactionation polymerase chain reaction (RTFQ-PCR). The expression of MUC2 protein was detected by immunocytochemistry and the protein expression of sucrase-isomaltase and lysozyme was detected by Western blot (Western blot). Results: aspirin could significantly inhibit the proliferation of HT-29 cells. The results of RT FQ-PCR and Western blot in a time and dose-dependent manner showed that aspirin inhibited the proliferation of HT-29 cells for 4872 hours, compared with the control group. The expression of m RNA in goblet cell marker MUC2TFF3 was up-regulated (P0.05), and the expression of m RNA and protein in intestinal absorption cell marker sucrase-isomaltase and lysozyme were down-regulated (P0.05). The expression of MUC2 protein was up-regulated at 48 h after aspirin intervention compared with the control group (P 0.05). Conclusion: aspirin can affect the expression of intestinal differentiation markers in human colon cancer HT-29 cells and may lead to the phenotype differentiation of goblet cells.
