龙抗Ⅰ号对放疗Lewis肺癌小鼠的减毒及增效作用的研究

【摘要】:恶性肿瘤是当前危害人类健康的重要疾病之一,每年有大量患者死于恶性肿瘤。放疗是治疗恶性肿瘤的三大手段之一,是利用放射线照射癌细胞,让其致死的疗法。放疗虽然能治疗肿瘤,但可引起患者的局部放射性损伤。因此在放疗的同时可以使用药物进行辅助治疗,一方面可以减轻放疗带来的毒副作用,另一方面可以增强放疗的治疗效果,达到减毒增效的作用。目前已有多种中医药已经用与辅助放疗。龙抗Ⅰ号取自我国传统的名贵中药,临床上应用十分广泛,与之有关的研究引起较多关注。龙抗Ⅰ号有活血化瘀、收敛止血、阵痛和抗炎的功效,并具有抗脂质过氧化,清除氧自由基,提高超氧化物歧化酶和过氧化氢酶的活性的重要生理作用。本课题组前期研究发现,龙抗Ⅰ号对于由重离子、中子、γ射线等辐射引起的大鼠损伤模型和由γ射线引起的骨髓型放射病小鼠模型以及γ射线引起的胃肠型放射病小鼠模型中,均具有一定的抗辐射作用,由此推断龙抗Ⅰ号具有潜在的辅助放疗的作用。在少量文献中报道过,龙抗Ⅰ号也具有一定的抗肿瘤作用。本研究继续探究龙抗Ⅰ号对肿瘤生长的影响以及对于放疗的减毒及增效作用。本研究使用C57/BL小鼠成功的建立了Lewis肺癌小鼠模型,然后以预防给药和治疗给药相结合的方式给予小鼠龙抗Ⅰ号,通过对小鼠一般生活状态、存活情况、体重变化、肿瘤体积和重量、肿瘤转移和血象水平等多方面的考察,研究龙抗Ⅰ号对Lewis肺癌小鼠的影响。用60Coγ射线对Lewis肺癌小鼠进行4Gy一次性全身辐射建立了放疗Lewis肺癌小鼠模型,并以预防给药和治疗给药相结合的方式对小鼠进行药物处理,通过观察小鼠的一般生活状态、体重变化、肿瘤体积和重量、肿瘤转移情况和血象水平,研究龙抗Ⅰ号对放疗Lewis肺癌小鼠的减毒及增效作用。同时进行了龙抗Ⅰ号对LLC细胞存活的影响的体外实验。实验结果表明,龙抗Ⅰ号对于Lewis肺癌小鼠和放疗Lewis肺癌小鼠的肿瘤生长和肿瘤转移没明显影响,但对Lewis肺癌小鼠和放疗Lewis肺癌小鼠存活情况、一般状态以及血象指标有一定改善。体内实验结果表明,在低剂量、短时间的处理条件下,龙抗Ⅰ号有一定的促进Lewis肺癌细胞存活的作用,但随着剂量以及时间的增加,龙抗Ⅰ号没有表现出促进或抑制Lewis肺癌细胞存活的作用,由于体内实验的给药相当于高浓度长时间的方式,所以体外实验与体内实验的结果一致,均可说明龙抗Ⅰ号没有促进Lewis肺癌细胞存活的作用。综合以上结果得出,龙抗Ⅰ号可以作为对放疗具有减毒作用的辅助用药。
[Abstract]:Malignant tumor is one of the most important diseases endangering human health. A large number of patients die of malignant tumor every year. Radiotherapy is one of the three methods to treat malignant tumor. Although radiotherapy can treat tumor, it can cause local radiation damage in patients. Therefore, the adjuvant therapy can be carried out with drugs while radiotherapy. On the one hand, it can reduce the toxic side effects brought by radiotherapy, on the other hand, it can enhance the therapeutic effect of radiotherapy and achieve the effect of reducing toxicity and increasing efficiency. At present, a variety of Chinese medicine has been used and adjuvant radiotherapy. Longkang I is taken from Chinese traditional Chinese medicine. It is widely used in clinic, and the related research has attracted more attention. Longkang I has the functions of promoting blood circulation and removing blood stasis, astringent hemostasis, labor pain and anti-inflammation. It also has the important physiological effects of anti-lipid peroxidation, scavenging oxygen free radicals and improving the activities of superoxide dismutase and catalase. Earlier studies by our research group have found that long Kang I is used by heavy ions, neutrons, and neutrons. The damage model of rats induced by 纬-ray isradiation, the model of bone marrow radiation sickness induced by 纬-ray and the model of gastrointestinal radiation sickness induced by 纬-ray have some anti-radiation effects.
