EGCG对胃癌细胞SHH信号通路Ptch、Gli-1的表达影响

【摘要】:目的:探讨(-)-Epigallocatechin gallate(EGCG)对胃癌细胞(AGS)增殖、凋亡影响,以及对Sonic hedgehog(SHH)信号通路相关蛋白Ptch、Gli-1的表达影响。方法:将AGS细胞株在37℃,5%CO2饱和湿度温箱里进行培养、换液和传代。实验组EGCG浓度为25μg/ml、50μg/ml、100μg/ml,并设置阴性对照组,实验时取对数期细胞。应用MTT法和流式细胞仪检测EGCG对AGS胃癌细胞增殖与凋亡,细胞爬片免疫组化检测SHH信号通路相关蛋白Ptch、Gli-1的表达影响。结果:EGCG干预AGS细胞48h后,随着EGCG浓度升高,细胞贴壁数量减少且形态变圆,漂浮数量增多,分布变稀疏,可见到细胞崩解后的碎片、颗粒等;而阴性对照组AGS细胞贴壁良好,轮廓清晰。随着EGCG浓度增高,AGE胃癌细胞增殖抑制率、凋亡率明显提高(P0.05);随着EGCG浓度的升高,Gli-1表达显著降低而Ptch表达显著升高(P0.05)。结论:EGCG可以抑制SHH信号通路的相关蛋白促使AGS胃癌细胞凋亡
