EHD2在非小细胞肺癌中的表达及其临床意义

【摘要】:目的研究EHD家族蛋白2(EHD2)在非小细胞肺癌(NSCLC)中的表达及其与患者临床病理特征和预后的关系。方法本研究采用蛋白印迹实验检测4对新鲜的NSCLC组织和癌旁组织中的表达,应用免疫组化法检测91例NSCLC组织中EHD2与Ki-67的表达,并分析其与临床参数之间的关系,及EHD2与NSCLC患者预后的关系。结果蛋白印迹实验显示EHD2在4对NSCLC癌组织中低表达,在癌旁组织中高表达;免疫组化结果表明EHD2的表达在分化好的癌组织中表达比分化差的组织高,其趋势与Ki-67的表达相反。NSCLC中EHD2与组织学分化、p TNM分期、肿瘤大小、淋巴结转移以及Ki-67密切相关;EHD2低表达与NSCLC患者的不良预后密切相关。结论 EHD2表达异常可能与NSCLC的发生发展关系密切;EHD2可作为判断NSCLC预后的指标,并可作为治疗的潜在靶点。
[Abstract]:Objective to investigate the expression of EHD family protein 2 (EHD2) in (NSCLC) and its relationship with clinicopathological features and prognosis of patients with NSCLC. Methods the expression of EHD2 and Ki-67 in 4 pairs of fresh NSCLC tissues and adjacent tissues was detected by Western blot. The expression of EHD2 and Ki-67 in 91 cases of NSCLC was detected by immunohistochemistry, and the relationship between the expression of EHD2 and clinical parameters was analyzed. And the relationship between EHD2 and prognosis of NSCLC patients. Results Western blot analysis showed that the expression of EHD2 was low in 4 pairs of NSCLC carcinomas and overexpressed in adjacent tissues. The results of immunohistochemistry showed that the expression of EHD2 was higher in well-differentiated tissues than in poorly differentiated tissues, and the trend was opposite to that of Ki-67. The expression of EHD2 in NSCLC was associated with histological differentiation of, p TNM stage and tumor size. Lymph node metastasis and Ki-67 were closely related. The low expression of EHD2 is closely related to the poor prognosis of NSCLC patients. Conclusion the abnormal expression of EHD2 may be closely related to the occurrence and development of NSCLC, and EHD2 can be used as a prognostic indicator of NSCLC and a potential target for treatment.
