端粒保护蛋白TRF2在肿瘤发生和治疗中作用机制的研究进展

【摘要】:端粒重复序列结合因子2(telomeric-repeat binding factor 2,TRF2)是一种重要的端粒保护蛋白,与端粒保护蛋白1(protection of telomeres 1,POT1)、端粒重复序列结合因子1(telomeric-repeat binding factor 1,TRF1)、相互作用核蛋白2(TRF1-interacting nuclear protein 2,TIN2)、阻滞活化蛋白1(repressor activator protein 1,Rap1)以及TPP1,5个核心蛋白通过一系列相互作用共同形成端粒保护蛋白复合体(shelterin)以维持端粒结构和功能的稳定性和完整性。越来越多的研究显示TRF2在多种肿瘤中异常表达并与肿瘤的发生,肿瘤细胞的耐药以及肿瘤血管的生成密切相关。因此,本文就TFR2的结构与生理功能,以及其在肿瘤发生,发展与治疗中的作用研究进展进行综述,以期为肿瘤的预防和治疗提供新的思路。
[Abstract]:Telomere repeat binding factor 2 (telomeric-repeat binding factor 2) is an important telomere protection protein associated with telomere protection protein 1 (protection of telomeres 1 (POT1), telomere repeat sequence binding factor 1 (telomeric-repeat binding factor 1) and telomere repeat sequence binding factor 1 (TRF1). Interaction nucleoprotein 2 (TRF1-interacting nuclear protein 2), block activator protein 1 (repressor activator protein 1), Rap1) and five core proteins of TPP1, form telomere protection protein complex (shelterin) through a series of interactions to maintain the stability and integrity of telomere structure and function. More and more studies show that the abnormal expression of TRF2 in many kinds of tumors is closely related to the occurrence of tumor, drug resistance of tumor cells and angiogenesis of tumor. In this paper, the structure and physiological function of TFR2 and its role in tumorigenesis, development and treatment are reviewed in order to provide new ideas for the prevention and treatment of tumor.
