白细胞介素-17与肝癌家族聚集性的关系

【摘要】:目的研究白细胞介素-17在广西肝癌高发地区肝癌高发家族成员与无癌家族成员血清中的表达情况,探讨IL-17的表达水平与广西肝癌家族聚集性的相关性。方法在广西肝癌高发地区配对选取112对肝癌高发家族和无癌家族成员,共224例,高发家族组有男65例,女47例,年龄(27.1±21.2)岁,无癌家族组有男65例,女47例,年龄(26.8±17.7)岁。112例高发家族成员中根据亲缘关系的不同,分一级亲属组、二级亲属组、三级亲属组、四级及以上亲属组,其中一级亲属62例,二级亲属11例,三级亲属25例,四级及以上亲属14例;根据高发家族组中患肝癌例数的不同,分2~3例组,4例及以上两组,其中2~3例组70例,4例及以上组42例。以高发家族成员为实验组,无癌家族成员为对照组,采用酶联免疫吸附(Enzyme-Linked Immunosorbent Assay,ELISA)法检测两组研究对象外周血清IL-17的表达水平。结果(1)高发家族组、无癌家族组IL-17的总体表达水平分别为(71.13±30.22)pg/ml、(19.01±14.46)pg/ml,高发家族成员IL-17表达水平显著高于无癌家族成员,差异有统计学意义(t=16.464,P=0.000);进一步分层分析:高发家族组男性、女性、年龄≤25岁、年龄25岁、HBs Ag阳性、HBs Ag阴性、壮族、瑶族成员IL-17的平均浓度分别为(71.16±28.43)pg/ml、(71.08±32.86)pg/ml、(69.32±29.53)pg/ml、(73.73±31.33)pg/ml、(70.14±20.11)pg/ml、(71.82±35.76)pg/ml、(72.38±33.65)pg/ml、(67.99±19.25)pg/ml,无癌家族组男性、女性、年龄≤25岁、年龄25岁、HBs Ag阳性、HBs Ag阴性、壮族、瑶族成员IL-17的平均浓度分别为(17.36±14.45)pg/ml、(21.30±14.30)pg/ml、(17.19±13.33)pg/ml、(21.63±15.71)pg/ml、(18.48±13.99)pg/ml、(19.39±14.87)pg/ml、(19.25±15.09)pg/ml、(18.41±12.93)pg/ml,两组比较,t值分别为13.603、9.524、13.072、10.081、14.307、10.999、12.886、12.094,所有的P值均0.05,高发家族组不同人口学特征IL-17表达水平都高于无癌家族组,差异有统计学意义。(2)高发家族组中不同亲缘系数的成员之间IL-17表达水平差异无统计学意义(Z=4.831,P=0.185)。(3)高发家族组中随着患肝癌例数增加,IL-17表达水平下降,但差异无统计学意义(t=0.580,P=0.563)。结论肝癌高发家族成员IL-17表达水平升高与肝癌家族聚集现象存在相关性。
