枸杞多糖在白血病患者中的应用及对HL-60和NK细胞杀伤活性的影响
[Abstract]:Aim to investigate the application of Lycium barbarum polysaccharides in patients with leukemia and the effect of Lycium barbarum polysaccharides on the expression of (MICA) protein on the surface of target cells on the surface of human anterior myeloid cells (HL-60) and the killing activity of natural killer (NK) cells and its mechanism. Methods HL-60 cells from leukemia patients were randomly divided into observation group 1, observation group 2 and observation 3 groups. The cells were co-cultured with Lycium barbarum polysaccharides at the concentration of 6,8,10 mg/ml, respectively. The expression of MICA protein on the surface of HL-60 cells from 30 leukemia patients without Lycium barbarum polysaccharide was measured by flow cytometry. The solution obtained from the four groups was mixed with NK cells for 20 hours, and the cytotoxicity of NK cells to four groups of HL-60 cells was determined by (MTT) assay with tetramethylazo blue colorimetric assay. Results the expression rate of MICA in group 1 was significantly lower than that in group 2, and in group 3 (P0.05), the expression rate of MICA in group 1, group 2 and group 3 was higher than that in control group (P0.05). With the increasing ratio of target cells and effector cells, the killing activity of NK cells in four groups increased, and the killing activity of NK cells in group 1, group 2 and group 3 was higher than that in control group (P0.05). The killing activity of NK cells in three groups was higher than that in observation group 1 and group 2 (P0.05). Conclusion HL-60 cells incubated with Lycium barbarum polysaccharide can increase the expression rate of MICA in leukemia patients, and there is a positive correlation between the concentrations. At the same time, Lycium barbarum polysaccharide can improve the killing activity of NK cells and clear the small lesions.
