DZNep通过下调EZH2蛋白表达抑制人前列腺癌PC3细胞增殖

【摘要】:目的研究EZH2蛋白抑制剂DZNep对前列腺癌PC3细胞增殖的影响及其用于人前列腺癌治疗的潜在价值。方法 MTT分析DZNep处理、siRNA敲低EZH2和EZH2过表达对PC3细胞增殖的影响,Western blot检测EZH2和Bax/Bcl-2的表达。结果 DZNep抑制PC-3细胞增殖、诱导细胞凋亡,上调Bax基因表达并下调Bcl-2基因表达。敲低EZH2抑制PC3细胞增殖,过表达EZH2则拮抗DZNep对PC3细胞的增殖抑制。结论 DZNep通过下调EZH2表达、诱导细胞凋亡发生从而抑制PC3细胞增殖。DZNep具有抗人前列腺癌治疗的潜在应用前景。
[Abstract]:Objective to investigate the effect of EZH2 protein inhibitor DZNep on proliferation of prostate cancer PC3 cells and its potential value in the treatment of human prostate cancer. Methods MTT was used to analyze the effects of DZNep treatment, siRNA knockdown EZH2 and EZH2 overexpression on the proliferation of PC3 cells., Western blot was used to detect the expression of EZH2 and Bax/Bcl-2. Results DZNep inhibited the proliferation of PC-3 cells, induced apoptosis, up-regulated the expression of Bax gene and down-regulated the expression of Bcl-2 gene. Low level of EZH2 inhibited the proliferation of PC3 cells, and over-expression of EZH2 inhibited the proliferation of PC3 cells induced by DZNep. Conclusion DZNep can inhibit the proliferation of PC3 cells by down-regulating the expression of EZH2 and inducing apoptosis. DZNep has the potential application prospect of anti-prostate cancer therapy.
