P53蛋白与miR375在青海地区胃癌患者血浆中的水平及临床意义

【摘要】：目的:探讨青海地区胃癌患者血浆中P53蛋白与mi R375的水平及临床意义,以了解两者是否可以联合作为预测胃癌发生的高特异性因子。方法:采用Elisa法及实时荧光定量PCR(RT-q PCR)检测35例胃癌患者(GC组)、35例慢性萎缩性胃炎患者(CAG组)及35例慢性非萎缩性胃炎者(CSG组)血浆中P53蛋白与mi R375的水平;观察不同疾病组患者P53蛋白与mi R375的水平变化情况。结果:1.血浆P53蛋白水平在CSG组、CAG组、GC组依次增高,分别为18.61±2.78、22.10±1.71、26.01±4.53(ng/ml)提示随着病变程度的加重,P53蛋白水平升高,三组间比较差异有统计学意义(P0.05)。2.血浆mi R375表达水平GC组较CAG组、CSG组明显降低,分别为1.41±0.56、2.76±0.39、2.75±0.21,与后两组比较差异有统计学意义(P0.01),而后两组之间差异无统计学意义(P=0.73)。3.在胃癌组中mi R375的表达量在不同肿瘤直径、淋巴结转移、分化程度、浸润深度、TNM分期间均有统计学差异(P0.05);P53蛋白水平在不同分化程度、不同浸润深度之间均有统计学差异(P0.05)。4.GC组血浆P53蛋白与mi R375间呈负相关(r=-0.575 P=0.026)。结论:1.与C SG组及C AG组相比,GC组中m i R375表达降低。而P53蛋白在C SG组、C AG组、G C组依次增高,提示病变程度的加重。2.联合检测血浆中p53蛋白与m i R375的水平可能对胃癌的诊断具有一定参考意义。
[Abstract]:Objective: to investigate the level and clinical significance of plasma p53 protein and mi R375 in patients with gastric cancer in Qinghai Province, and to explore whether the combination of p53 protein and R375 can be used as a high specific factor to predict the occurrence of gastric cancer. Methods: 35 patients with gastric cancer (GC group) were detected by Elisa and real-time fluorescence quantitative PCR (RT-q PCR). The levels of p53 protein and mi R375 in plasma of 35 patients with chronic atrophic gastritis (CAG group) and 35 patients with chronic non-atrophic gastritis (CSG group). The changes of p53 protein and mi R375 in different disease groups were observed. Result: 1. The plasma p53 protein levels in CSG group, CAG group and GC group were 18.61 卤2.78, 22.10 卤1.71 and 26.01 卤4.53 (ng/ml), respectively, suggesting that the level of p53 protein increased with the severity of the lesion. There was significant difference among the three groups (P 0.05). 2. The expression level of plasma mi R375 in GC group was significantly lower than that in CAG group and CSG group, which was 1.41 卤0.56, 2.76 卤0.39 and 2.75 卤0.21, respectively, which was significantly different from that in the latter two groups (P 0.01).
