自发性乳腺肿瘤C3H/HeJNju小鼠原发肿瘤组织与脾脏间CD4 + CD25 + Treg TCR β CDR3组库异

目的:采用Illumina Solexa高通量测序技术,分析自发性乳腺肿瘤C3H/HeJNju小鼠原发乳腺肿瘤组织与脾脏间CD4+CD25+TregTCRβ CDR3组库的同质性与异质性,探讨肿瘤组织与和全身免疫循环之间CD4+CD25+Treg的区别和联系,为进一步研究乳腺癌中Treg的机制与应用提供基础。方法:1.10月龄以上的自发性乳腺肿瘤模型C3H/HeJNju出瘤小鼠3只,分别标记为M1、M2、M3。2.分别采集M1、M2、M3小鼠原发乳腺肿瘤组织(T)和脾脏(S),采用磁珠分选技术分选出各组织CD4+CD25+Treg细胞,FCM鉴定纯度。3.提取各分选获得的CD4+CD25+Treg细胞基因组DNA,样本序号命名:M1T:M1小鼠乳腺肿瘤组织样本;M1S:M1小鼠脾脏样本;M2T:M2小鼠乳腺肿瘤组织样本;M2S:M2小鼠脾脏样本;M3T:M3小鼠乳腺肿瘤组织样本;M3S:M3小鼠脾脏样本。4.基因组DNA样本送Adaptive Biotechnologies Immuno SEQ公司(美国华盛顿医学院),经检测符合建库要求后,按小鼠TCR β CDR3组库制备技术建立每...
