协同共刺激分子B7-H3对胃癌侵袭转移的影响及机制研究

目的1.检测胃癌标本中B7-H3的表达,分析B7-H3表达与胃癌患者临床病理特征之间的关系。2.探究B7-H3对胃癌生物学功能的影响。3.初步探究B7-H3与CXCR-4相互作用机制。方法1.收集120例苏州大学附属第二医院2014-2015年,未进行放化疗的手术切除的胃癌患者标本。免疫组化检测患者肿瘤组织中B7-H3的表达情况,并统计学分析B7-H3表达和临床资料的相关性。2.运用慢病毒感染人胃癌N87细胞株,下调其B7-H3的表达,并通过荧光显微镜观察、流式检测(flow cytometry,FCM)、免疫印迹试验(western blot)、实时定量PCR(real-time PCR)的方法验证感染率和感染效率。3.通过CCK-8、划痕、transwell实验分别检测B7-H3对胃癌细胞增殖能力、迁移能力、侵袭转移能力的影响。4.利用balb/c小鼠建立肿瘤转移模型,通过3D小动物成像观察小鼠主要脏器的肿瘤转移情况。5周终止实验,取出小鼠主要脏器,通过肉眼观察和HE染色观察肿瘤转移情况,进一步研究B7-H3在体内对肿瘤侵袭转移能力的影响。5.通过免疫组化检测胃癌患者肿瘤组织中肿瘤...
