【摘要】:目的拟构建气虚痰浊证高血压大鼠模型,为中医药研究提供载体。方法将50只12周龄自发性高血压大鼠(spontaneously hypertensive rats,SHRs),随机分为模型组(40只)及对照组(10只),同时设置10只12周龄正常血压大鼠(wistar-Kyoto,WKY)为对照(WKY组)。模型组隔日喂食高脂饲料,每周6次力竭游泳;对照组及WKY组始终以普通大鼠饲料喂养。对各组大鼠表征体系进行动态观察,12周后进行相关指标检测。结果模型组大鼠随着造模时间延长,分别出现生物学表征评分升高、游泳时间缩短、体质量增加、旷场分析中央格停留时间延长、水平运动距离缩短、血压升高等;血清指标检测中,模型组甘油三酯(TC)、胆固醇(TG)、血糖(GLU)均高于其他2组(P0.01);模型组低密度脂蛋白(LDL-C)较WKY组升高(P0.05);模型组双侧颈动脉血流量比对照组、WKY组降低,差异有统计学意义(P0.01)。结论以SHRs为基础,持续力竭游泳、隔日禁食、高脂饲料喂养建立气虚痰浊证高血压大鼠模型,相关表征及血清指标改变符合气虚痰浊证候特点。
[Abstract]:Objective to construct the model of hypertension rats with Qi deficiency and phlegm turbidity syndrome, and to provide the carrier for the research of traditional Chinese medicine. Methods a total of 50 12 week old spontaneously hypertensive rats (spontaneously hypertensive rats, SHRs) were randomly divided into model group (40 rats) and control group (10 rats), and 10 12 week old normotensive rats (wistar-Kyoto, WKY) were served as control group (WKY group). The model group fed high fat diet every other day, swimming 6 times a week, and the control group and the WKY group were always fed with the common rat feed. The dynamic observation was carried out in each group of rats, and the related indexes were detected after 12 weeks. The rats in the model group with the modeling time, respectively. The biological characterization score was increased significantly, swimming time, increased body weight, open field test, the central grille time, horizontal movement distance shortened, high blood pressure; serum marker, glycerol three in the model group (TC) and cholesterol ester (TG), blood glucose (GLU) were higher than the other 2 groups (P0.01); model group, low density lipoprotein (LDL-C) was higher than that in WKY group (P0.05 group); carotid artery blood flow model than the control group, WKY group decreased, the difference was statistically significant (P0.01). Conclusion based on SHRs, a rat model of qi deficiency, phlegm turbid syndrome and hypertension is established based on continuous exhaustive swimming, fasting and high-fat diet. The related characteristics and serum indexes are in accordance with the characteristics of qi deficiency, phlegm turbidity syndrome.
