【摘要】:目的:通过比较寻常型银屑病血热证患者外周血中Th17、Treg细胞的特异性转录因子RORγt、Foxp3mRNA的表达与健康对照组的差异,分析其与血热证证型的相关性,以期为寻常型银屑病血热证的微观辨证做一些补充性研究。方法:采用实时荧光定量逆转录聚合酶链反应(real-time RT-PCR),对寻常型银屑病血热证患者外周血中Th17、Treg细胞的特异性转录因子RORγt、 Foxp3mRNA的表达水平进行量化分析。结果:1、寻常型银屑病血热证组30例外周血中Th17、Treg细胞的特异性转录因子RORγt, Foxp3mRNA相对表达量分别为1.99±0.81,1.13±0.56,分别与正常对照组相比,差异有统计学意义(t值分别为2.50,2.57,P均0.05),说明患病组外周血中RORγtmRNA的表达量明显高于对照组,Foxp3mRNA的表达量则明显低于对照组;2、寻常型银屑病血热证组30例外周血中RORγt/Foxp3表达量的比值为1.04±0.77,与正常对照组相比,差异无统计学意义(P0.05)。结论:1、寻常型银屑病血热证患者外周血中Th17细胞转录因子RORγtmRNA与健康人相比较呈高表达,可以佐证Th17细胞参与了寻常型银屑病血热证证候的产生;寻常型银屑病血热证患者外周血中Treg细胞转录因子Foxp3mRNA与健康人相比呈低表达,可以佐证Treg细胞参与了寻常型银屑病血热证证候的产生。但转录因子RORγ、Foxp3仅仅是Th17、Treg细胞参与发病的一小部分,从多层次、多方位研究Th17、Treg细胞与寻常型银屑病血热证证候相关性需要我们进一步去探索;2、寻常型银屑病血热证患者转录因子RORγt/Foxp3表达与健康人比较没有统计学意义,但近几年很多资料证实Th17/Treg细胞失衡是寻常型银屑病发病的关键,RORγt是Th17细胞转录因子,Foxp3是Treg细胞转录因子,RORγt/Foxp3表达比值不能完全反映Th17/Treg细胞失衡的情况。
[Abstract]:Objective: to compare the expression of ROR 纬 ttcr3 mRNA in peripheral blood of patients with blood fever of psoriasis vulgaris. Analysis of its correlation with the type of blood heat syndrome. Objective: to do some supplementary research on micro differentiation of blood fever syndrome of psoriasis vulgaris. Methods: Real-time RT PCR was used to detect Psoriasis vulgaris. The expression of ROR 纬, Foxp3mRNA in peripheral blood of patients with psoriasis vulgaris was quantitatively analyzed. The relative expression of ROR 纬 t and Foxp3mRNA in peripheral blood of 30 patients with blood fever of psoriasis vulgaris was 1.99 卤0.81, respectively. 1.13 卤0.56, compared with the normal control group, the difference was statistically significant (2.50 卤2.57g, P < 0.05). The results showed that the expression of ROR 纬 tmRNA in the peripheral blood of the diseased group was significantly higher than that of the control group, and the expression of Foxp3 mRNA was significantly lower than that of the control group. 2, the ratio of ROR 纬 t / Foxp3 expression in peripheral blood of 30 patients with psoriasis vulgaris was 1.04 卤0.77, compared with that of normal controls. Conclusion: Th17 transcription factor ROR 纬 tmRNA in peripheral blood of patients with blood fever of psoriasis vulgaris is higher than that of healthy persons. It can be proved that Th17 cells are involved in the production of blood fever syndrome of psoriasis vulgaris. The expression of Treg transcription factor Foxp3mRNA in peripheral blood of patients with blood fever of psoriasis vulgaris was lower than that of healthy people. It can be concluded that Treg cells are involved in the production of blood fever syndrome of psoriasis vulgaris, but the transcription factor ROR 纬 Foxp3 is only a small part of Th17T Treg cells involved in the pathogenesis of psoriasis vulgaris. The relationship between Th17 Treg cells and blood fever syndrome of psoriasis vulgaris needs to be further explored. 2. The expression of transcription factor ROR 纬 t / Foxp3 in patients with blood fever of psoriasis vulgaris was not significantly different from that in healthy persons. However, in recent years, many data have confirmed that Th17/Treg cell imbalance is the key to the pathogenesis of psoriasis vulgaris. ROR 纬 t is a transcription factor of Th17 cells and Foxp3 is a transcription factor of Treg cells. The ratio of ROR 纬 t / Foxp3 expression can not completely reflect the imbalance of Th17/Treg cells.
