【摘要】:目的:采用体外细胞实验及小鼠模型实验来研究阿里红多糖抗氧化、抗肿瘤及免疫调节活性,为多糖进一步研究提供一定的实验依据。方法:1、选择体外清除自由基反应体系,采用二苯代苦味脐基自由基(DPPH·)、邻苯三酚自氧化法、羟基自由基(·OH)法等方法对考察阿里红多糖的体外抗氧化能力。2.采用四甲基偶氮哇蓝(MTT)显色法,观察不同浓度的阿里红多糖对鼠源乳腺癌4TI细胞、人结肠癌细胞系HT-29、人宫颈癌细胞系Hela、人卵巢癌细胞系SK-OV-3、人前列腺癌细胞系PC-3、人肝癌细胞系Hep3B细胞生长的抑制作用,筛选阿里红多糖抗肿瘤活性。3.采用ICR小鼠建立4T1荷瘤小鼠模型,观察阿里红多糖对模型小鼠移植性肿瘤的抑制作用影响。4.采用RSB法考察对小鼠脾脏T、B淋巴细胞的影响;探讨其体外免疫作用。结果:1、体外抗氧化结果显示阿里红多糖在体外3种抗氧化体系中均显示具有抗氧化活性。2、在体外,各剂量浓度的阿里红多糖对鼠源乳腺癌4Tl细胞增殖有抑制作用。3、阿里红多糖能有效抑制实体瘤模型荷瘤小鼠体内肿瘤生长,结果显示阿里红多糖对4T1小鼠的抑瘤率分别为58.69%(低剂量组)、60.89%(中剂量组)、74.73%(高剂量组)。4、阿里红多糖对脾细胞具有免疫调节作用。FoP既能与ConA或LPS协同诱导小鼠脾T、B淋巴细胞的增殖,也能直接促进小鼠脾T、B淋巴细胞的增殖;FoP也可提高小鼠脾细胞CD19+CD86+B细胞的比值;此外,FoP还可促进T淋巴细胞分泌Th1型细胞因子IL-2分泌和B细胞活化和增殖的IL-13的分泌,对主要来源于巨噬细胞的细胞因子TNF-α的分泌也有促进作用。5、FoP也能诱导小鼠腹腔巨噬细胞的增殖,能促使来源于巨噬细胞的细胞因子TNF-α分泌。结论:1.阿里红多糖体外具有抗氧化活性。2.阿里红多糖在体内外具有抑制鼠源乳腺癌细胞株4T1的细胞增殖作用。3、能够显著促进正常小鼠脾淋巴细胞增殖,且在一定范围内具有剂量依赖关系,具有免疫作用。
[Abstract]:Objective: to study the anti-oxidation, anti-tumor and immunomodulatory activities of Ari Red polysaccharides in vitro and mouse model experiments, and to provide some experimental basis for further study of polysaccharides. Methods: 1. The free radical scavenging reaction system in vitro was selected and the diphenyl bitter umbilical radical DPPH 路N and pyrogallol autoxidation were used. Hydroxyl radical (路OH) method was used to study the antioxidation ability of Ari Red polysaccharides in vitro. 2. Tetramethylazo blue blue (MTT) method was used. The effects of different concentrations of Ali Red polysaccharides on mouse breast cancer cell line 4TI, human colon cancer cell line HT-29, human cervical cancer cell line Hela and human ovarian cancer cell line SK-OV-3 were observed. The growth inhibition of human prostate cancer cell line PC-3 and human hepatoma cell line Hep3B was studied. The anti-tumor activity of Ali Red polysaccharide was screened. The model of 4T1 tumor-bearing mice was established by using ICR mice. To observe the inhibitory effect of Ali Red Polysaccharide on transplanted tumor in model mice. (4) to investigate the effect of RSB on spleen TnB lymphocytes in mice; Results the results showed that the polysaccharides had antioxidant activity of 0.2 in vitro and showed antioxidant activity in vitro. Different doses of Ali Red polysaccharides can inhibit the proliferation of mouse breast cancer 4TL cells. 3. Ali red polysaccharides can effectively inhibit the tumor growth of solid tumor model mice. The results showed that the inhibitory rates of the polysaccharide against 4T1 mice were 58.69 (low dose group 60.89) (middle dose group 74.73%) (high dose group 74.73%). Ali Red polysaccharides have immunomodulatory effect on spleen cells. Fop can not only induce the proliferation of Thyrosine B lymphocytes in mice spleen in cooperation with ConA or LPS, but also directly promote the proliferation of Thyrosine B lymphocytes in mice spleen. FoP also increased the ratio of CD19 CD86 B cells in spleen cells of mice. In addition, FoP can also promote T lymphocytes to secrete Th1 type cytokine IL-2 secretion and B cell activation and proliferation of IL-13 secretion. It also promoted the secretion of TNF- 伪, a cytokine mainly derived from macrophages, and also induced the proliferation of murine peritoneal macrophages. TNF- 伪, a cytokine derived from macrophages, can be secreted. Conclusion:. 1. Ali red polysaccharide has antioxidant activity in vitro. In vitro and in vivo, Ali red polysaccharide can inhibit the proliferation of mouse breast cancer cell line 4T1. 3. It can significantly promote the proliferation of splenic lymphocytes in normal mice, and has a dose-dependent relationship in a certain range, and has immune effect.
