穿心莲内酯对病毒性心肌炎小鼠保护作用及其机制的研究

【摘要】：背景与目的病毒性心肌炎(Viral Myocarditis,VMC)是病毒侵犯心脏引起的以心肌细胞的变性坏死和心肌间质炎症为主的一种疾病。VMC的发病机制目前仍不十分清楚,治疗上缺乏有效而特异的手段。本研究通过柯萨奇病毒B3(Coxsackievirus B3,CVB3)感染Balb/c小鼠,建立小鼠VMC模型,并采用穿心莲内酯注射液进行治疗干预,观察穿心莲内酯对病毒性心肌炎小鼠模型的作用并探讨其可能机制。方法1.150只雄性Balb/c小鼠随机分为正常对照组(对照组,n=40),病毒感染组(感染组,n=60),穿心莲内酯治疗组(治疗组n=50)。2.感染组与治疗组腹腔接种0.2ml含CVB3的病毒液,对照组注射等体积的生理盐水。治疗组0.5h后腹腔注射0.2ml 125mg/kg.d穿心莲内酯注射液,连续用药2周,对照组与感染组每天注射等体积的生理盐水。3.观察小鼠生存状态,统计小鼠死亡率,于病毒接种后7d和14d观察心肌组织病理学改变、血清白细胞介素6(IL-6)水平、心肌组织中肿瘤坏死因子α(TNF-α)表达水平。结果1.小鼠接种病毒第7天及14天,与对照组比较,感染组小鼠血清IL-6、心肌TNF-α表达、心肌病理评分显著增加,差异有统计学意义(P0.05)。2.小鼠病毒接种后7天及14天,与感染组相比,治疗组小鼠血清IL-6、心肌TNF-α表达、心肌病理评分降低,差异有统计学意义(P0.05)。结论穿心莲内酯能够减轻病毒性心肌炎小鼠心肌损害。其机制可能抑制血清IL-6、心肌TNF-α表达水平,减轻心肌炎心肌损伤。
[Abstract]:Background and objective virus Myocarditis in viral myocarditis. VMC is a kind of disease caused by virus invading the heart. The pathogenesis of VMC is not clear at present. It is mainly caused by degeneration and necrosis of myocardial cells and interstitial inflammation of myocardium. In this study, Balb/c mice were infected with Coxsackievirus B3Coxsackievirus B3CVB3. Mouse VMC model was established and treated with andrographolide injection. To observe the effect of andrographolide on viral myocarditis in mice and explore its possible mechanism 1.150 male Balb/c mice were randomly divided into normal control group (control group 40). Virus infection group (infection group), andrographis lactone treatment group (treatment group) (nonglactide treatment group). The infection group and the treatment group were intraperitoneally inoculated with 0.2 ml of virus solution containing CVB3. The control group was injected with the same volume of normal saline, and the treatment group was injected with 0.2 ml 125mg / kg 路d andrographolide injection for 2 weeks after 0.5 h. Control group and infection group were injected with saline of the same volume every day. The survival status of mice was observed and the mortality of mice was counted. The pathological changes of myocardium were observed 7 and 14 days after inoculation. The level of serum interleukin-6 (IL-6) and the expression of TNF- 伪 in myocardial tissue. Results 1.The mice were inoculated with the virus on the 7th and 14th day, compared with the control group. Serum IL-6, TNF- 伪 expression and cardiomyopathology score were significantly increased in the infected group. The difference was statistically significant (P0.05. 2.The mice were inoculated 7 and 14 days after inoculation). Compared with the infection group, the serum IL-6, TNF- 伪 expression and cardiomyopathology score in the treatment group were lower than those in the control group. Conclusion andrographolide can reduce myocardial damage in mice with viral myocarditis and its mechanism may inhibit the expression of serum IL-6 and myocardial TNF- 伪. Reduce myocardial injury in myocarditis.
