基于网络药理学的沙棘总黄酮治疗心肌缺血的作用机制研究

【摘要】:该研究采用网络药理学虚拟筛选方法,旨在进一步研究沙棘总黄酮(total flavonoids of Hippophae rhamnoides,TFH)治疗心肌缺血的活性成分和作用机制。首先通过TCMSP数据库和PubChem数据库,结合口服利用度和类药性分析,筛选出沙棘化合物中的黄酮类化合物。筛选得到的7个化合物再通过Chem Mapper服务器进行靶点的预测分析。将得到的潜在靶点导入MAS 3.0数据库,结合KEGG数据库进行靶点分析和通路分析。最后使用Cystoscope 3.3.0软件绘制"化合物-靶点-作用通路"网络图。虚拟实验预测得到了68个潜在靶点和60条信号通路,其中有31个靶点和23条通路直接或间接与心肌缺血有关。结果表明,TFH主要通过对钙离子信号通路(calcium signaling pathway)、血管内皮生长因子信号通路(VEGF signaling pathway)和缝隙连接信号通路(gap junction)等信号通路的调节发挥协同作用,且与文献报道吻合;提示其可能通过参与调节血小板聚集、脂质代谢、炎症反应等过程,增强心功能和对血管内皮细胞保护作用,从而发挥抗心肌缺血的作用。
[Abstract]:The virtual screening method of network pharmacology was used in this study. Objective to further study the total flavonoids of Hippophae rhamnoides of Hippophae rhamnoides. The active components and the mechanism of action of TFH in the treatment of myocardial ischemia were analyzed by TCMSP database and PubChem database combined with oral availability and drug-like analysis. The flavonoids from Hippophae rhamnoides compounds were screened. 7 compounds were screened and analyzed by Chem Mapper server. The potential targets were imported into MAS. 3.0 Database. In combination with KEGG database, target analysis and path analysis are carried out. Finally, "Compound-target-Action Pathway" is drawn by using Cystoscope 3.3.0 software. Network Diagram. Virtual experiment predicts 68 potential targets and 60 signal pathways. Among them, 31 targets and 23 pathways were directly or indirectly related to myocardial ischemia. TFH mainly through calcium signaling pathway. Vascular endothelial growth factor (VEGF signaling pathway) and gap junction signal pathway (gap junction). The regulation of iso-signal pathway plays a synergistic role. The results were in good agreement with the literature reports. These results suggest that it may play a role in anti-myocardial ischemia by regulating platelet aggregation, lipid metabolism and inflammatory reaction, enhancing cardiac function and protecting vascular endothelial cells.
