瑶药铜钻总黄酮对急性心肌缺血再灌注损伤保护作用的研究

【摘要】:目的:研究瑶药铜钻(Mappianthus iodoies Hand-Mazz)总黄酮对急性心肌缺血再灌注损伤的保护作用。方法:以72只成年健康雄性SD大鼠作为研究对象,随机将其平均分为假手术组(Sham)、再灌注模型组(Model)、阳性药物对照组(YXDZ)、瑶药铜钻总黄酮低剂量组(YYTZ-L 60 mg·kg-1)、瑶药铜钻总黄酮中剂量组(YYTZ-M 120 mg·kg-1)和瑶药铜钻总黄酮高剂量组(YYTZ-H 240 mg·kg-1)六个组。结扎大鼠左冠状动脉前降支造成缺血30 min、解除结扎形成再灌注24 h的方法建立缺血再灌注模型。通过HE染色法对大鼠心肌组织病理形态学改变进行观察;用TTC染色法检测大鼠心肌梗死面积;ELISA试剂盒检测大鼠血清中超氧化物歧化酶(SOD)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、肌酸激酶(CK)、乳酸脱氢酶(LDH)的浓度;免疫印迹检测B细胞淋巴瘤/白血病-2(BCL-2)和BCL-2相关X蛋白(BAX)蛋白表达。结果:与Model相比,瑶药铜钻总黄酮剂量组对大鼠急性心肌缺血再灌注损伤的心肌组织病理形态学损害均有明显改善,心肌梗死面积明显较小(P0.05),血清中MDA、TNF-α、IL-6、CK、LDH含量均较低(P0.05),SOD含量较高(P0.05),BCL-2蛋白水平表达升高、BAX蛋白水平表达降低(P0.05);与Sham相比,Model心肌组织病理形态学损伤较明显,心肌梗死面积明显较大(P0.05),大鼠血清中CK、LDH、MDA、TNF-α、IL-6含量均明显增高(P0.05),SOD含量较低(P0.05),大鼠BCL-2蛋白水平表达降低、BAX蛋白水平表达升高(P0.05)。结论:瑶药铜钻总黄酮对急性心肌缺血再灌注损伤有一定的保护作用,其机制可能与其抗炎、抗氧化及对抑制细胞凋亡作用有关。
[Abstract]:Objective: to study the iodoies Hand-Mazzs of Chinese herbal medicine Mappianthus iodoies Hand-Mazz. The protective effects of total flavonoids on acute myocardial ischemia-reperfusion injury were studied in 72 adult male SD rats. They were randomly divided into sham-operated group, reperfusion model group, and positive drug control group (YXDZ). YYTZ-L 60 mg 路kg-1). YYTZ-M (120 mg 路kg-1) and YYTZ-H (240 mg 路kg -1). The left anterior descending coronary artery was ligated for 30 min. The ischemia reperfusion model was established by removing ligation and forming reperfusion for 24 h. The pathological changes of myocardium in rats were observed by HE staining. Myocardial infarction area was measured by TTC staining. Serum superoxide dismutase (SOD), malondialdehyde (MDA), tumor necrosis factor- 伪 (TNF- 伪), interleukin-6 (IL-6) were detected by ELISA kit. The concentration of creatine kinase (CK) and lactate dehydrogenase (LDH); The expression of B-cell lymphoma / Leukemia BCL-2) and BCL-2 associated X protein was detected by Western blotting. Results: compared with Model. The total flavonoid dosage group of Yao medicine copper drill significantly improved the myocardial histopathologic damage of acute myocardial ischemia-reperfusion injury in rats, the myocardial infarction area was significantly smaller than that of P0.05, and MDA in serum was also significantly improved. The content of TNF- 伪, IL-6, CKL and LDH was higher than that of P0.05 and SOD, and the expression of BCL-2 protein was higher than that of P0.05 and BCL-2. The expression of BAX protein decreased in P0.05; Compared with Sham, myocardial histopathologic damage was more obvious, myocardial infarction area was significantly larger than P0.05, and serum CKD LDHDA-TNF- 伪 was found in rats. The content of IL-6 was significantly higher than that of P0.05, and the expression of BCL-2 protein was decreased in rats. Conclusion: the total flavonoids of Chinese herbal medicine have protective effect on acute myocardial ischemia-reperfusion injury, and its mechanism may be related to its anti-inflammatory mechanism.
