不同腧穴配伍电针防治大鼠心肌缺血损伤作用的比较

【摘要】:目的:比较不同腧穴配伍电针对心肌缺血大鼠针效的差异,为临床防治心肌缺血的选穴配伍提供依据。方法:Wistar雄性大鼠随机分为正常组、模型组、标本配穴组(双"内关"配伍双"足三里""关元")、常规配穴组(双"内关"配伍双"神门""膻中"),每组各10只。盐酸异丙肾上腺素(5mL/kg)皮下注射法复制心肌缺血模型。标本配穴组和常规配穴组在每日造模前电针处理10min,造模7d结束后仍每日电针,连续21d。检测各组大鼠心电图,采用全自动生化仪检测血清肌钙蛋白T(cTnT)水平,酶联免疫法检测心肌组织丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,透射电镜观察心肌细胞超微结构变化。结果:与正常组比较,模型组心电图S-T段振幅、心率(HR),血清cTnT和心肌组织MDA含量均显著升高(P0.01),心肌组织SOD和GSH-Px活性显著下降(P0.01),心肌超微结构破坏明显,线粒体肿胀变形,甚至空泡化;电针干预后,两电针组心电图S-T段振幅、HR,血清cTnT和心肌组织MDA含量显著下降(P0.05),心肌组织SOD和GSH-Px活性显著升高(P0.05),心肌超微结构损伤减轻;两电针组组间比较,标本配穴组血清cTnT和心肌组织MDA含量明显低于常规配穴组(P0.05),心肌组织SOD和GSH-Px活性明显高于常规配穴组(P0.05),标本配穴组心肌超微结构损伤程度较常规配穴组轻。结论:电针能有效防治心肌缺血,标本配穴法电针的防治作用优于常规配穴。
[Abstract]:Objective: to compare the acupuncture effect of different acupoints combined with electroacupuncture on myocardial ischemia rats. Methods: male Wistar rats were randomly divided into normal group and model group. The specimens were divided into two groups (double "Neiguan" and "Zusanli" and "Guanyuan") and conventional acupoint group (double "Neiguan" and "Shenmen" and "Shanzhong"). Myocardial ischemia model was established by subcutaneous injection of isoproterenol hydrochloride (5 mL / kg) in each group. The electrocardiogram (ECG) of the rats in each group was measured and the serum troponin TnTnT level was measured by automatic biochemical instrument after 7 days of modeling. The content of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in myocardium were detected by enzyme-linked immunosorbent assay (Elisa). Results: compared with the normal group, the amplitude of S-T segment and heart rate of the model group were compared with those of the normal group. The contents of serum cTnT and myocardial MDA increased significantly (P 0.01), and the activities of SOD and GSH-Px decreased significantly (P 0.01). The ultrastructure of myocardium was destroyed obviously. Mitochondria swelling and deformation, even vacuolation; After electroacupuncture intervention, the amplitudes of S-T segment of electrocardiogram (S-T) and the contents of serum cTnT and myocardial MDA in the two electroacupuncture groups decreased significantly (P 0.05). The activities of SOD and GSH-Px in myocardial tissue increased significantly (P 0.05), and the myocardial ultrastructure injury was alleviated. The contents of serum cTnT and myocardial MDA were significantly lower in group A than those in group B (P 0.05). The activities of SOD and GSH-Px in myocardial tissue were significantly higher than those in the control group (P0.05). Conclusion: electroacupuncture can effectively prevent and treat myocardial ischemia.
