针刺对术后结肠ICCs修复微环境中CSE表达的影响

【摘要】:研究目的通过动物实验,本研究观察针刺“足三针”穴组对大鼠腹部术后结肠Cajal细胞修复微环境中CSE表达的影响,深入探究针刺对胃肠动力障碍的影响和机制研究,并进一步深入了解针灸在腹部术后胃肠动力障碍功能的恢复方面提供了一定的理论依据。研究方法在相同实验条件下,将30只SPF级成年健康SD大鼠随机分为10只正常组、10只模型组、10只针刺组。取模型组和针刺组大鼠,术前禁食12h,10%水合氯醛行腹腔注射麻醉(按照330ml/kg),实验大鼠实施结肠吻合术,做成胃肠动力障碍结肠术后模型。针刺组予术后次日起每日给予针刺足三针穴组,模型组和针刺组保持同时进行,每天同一时间行空抓15min,来降低人为固定的影响因素。正常组不予处理。连续干预3天。期间,记录好每天各老鼠的进食量、大便颗粒和风干后大便的质量。实验结束后,观察小肠推进率并取各组大鼠结肠(盲肠后约2cm处),将取出的组织称量、标记、记录质量,尽快放入-80℃冰箱中保存,随后进行CSE表达的检测。研究结果1.测定术后大鼠胃肠道排空30min后小肠推进率的情况,与正常组相比,模型组术后小肠推进率较正常组显著下降(P<0.05),针刺组比模型组小肠推进率显著增加,其差异具有统计学意义(P<0.05)。实验表明针刺对腹部术后大鼠胃肠动力障碍具有改善作用。2.结肠CSE活性在术后有了大幅度的提升,模型组大鼠结肠CSE活性较正常组显著升高(P<0.05)。与模型组相比,针刺组CSE活性明显降低,且差异具有统计学意义(P<0.05);其结果说明针刺治疗能降低结肠组织中CSE的活性。通过针刺“足三针”穴组,腹部术后大鼠结肠ICCs修复微环境中CSE活性显著降低。研究结论针刺“足三针”穴组能明显改善腹部术后大鼠的胃肠功能障碍,调整大鼠腹部术后结肠Cajal细胞修复微环境中CSE的活性,从而改善胃肠功能障碍的症状。
