穴位注射和中药茶对去势大鼠内分泌和SIRT1水平的影响

【摘要】:目的:观察穴位注射和中药茶对去势大鼠内分泌和沉默信息调节因子2相关酶1(SIRT1)水平的影响,以探讨其治疗更年期综合征、延缓更年期衰老的作用机制。方法:选取3月龄雌性未孕SD大鼠156只,体重(250g±20g),为了更贴切地模拟人类更年期卵巢渐衰的过程,将其随机分为假手术组以及三大组群:次切组群、次全切组群和全切组群(每个组群里分别包含:模型组、中药茶组、穴位注射组和中药茶+穴位注射组四个亚组),共13组,每组各12只大鼠。运用次切、次全切、全切三种去势手术方法造模,次切:右侧卵巢全部切除,左侧卵巢只切除约50%;次全切:右侧卵巢全部切除,左侧卵巢只切除约80%;全切:双侧卵巢全部切除。成模后,中药茶组自由饮药,共饮用8周。穴位注射组选取关元、归来(双)、肝俞(双)、肾俞(双),穴位注射丹红注射液0.02mL/穴,1周1次,共8次。末次治疗结束后,采用ELISA法检测各组大鼠血清雌二醇(E2)、卵泡刺激素(FSH)以及心脏组织SIRT1蛋白的水平;取大鼠子宫组织HE染色后观察其形态结构的变化。结果:与假手术组相比,各模型组大鼠血清E2水平和心脏组织SIRT1蛋白的水平均显著降低,血清FSH的水平显著升高,其差异均具有统计学意义(P0.05);与模型组相比,各治疗组大鼠血清E2水平和心脏组织SIRT1蛋白的水平均显著升高,血清FSH的水平均显著降低,其差异均具有统计学意义(P0.05);其中,次切中药茶+穴位注射组大鼠的E2水平和SIRT1蛋白水平均值最高,FSH水平均值最低。大鼠子宫内膜形态学观察结果提示:各治疗组均能使大鼠子宫内膜上皮增厚,使间质致密、腺体增多、血管丰富,其中,以次切中药茶+穴位注射组治疗效果最佳。结论:(1)本实验运用次切、次全切、全切三种去势手术方法造模,成功地复制了更年期大鼠模型,并模拟了人类更年期卵巢渐衰的过程。(2)穴位注射和中药茶能延缓去势大鼠衰老进程,其作用机制可能是穴位注射和中药茶能提高去势大鼠血清E2水平以及心脏组织SIRT1蛋白的水平,降低血清FSH水平。(3)各治疗组间的数据结果提示:在大鼠卵巢渐衰的过程中,早期联合干预治疗疗效更佳。
[Abstract]:Objective: to observe the effects of acupoint injection and traditional Chinese medicine tea on endocrine and silent information regulatory factor-2 related enzyme 1SIRT1 in ovariectomized rats, in order to explore the treatment of climacteric syndrome. Methods: 156 3-month-old female non-pregnant SD rats, weighing 250g 卤20g / g, were selected to simulate the process of ovarian decline in human menopause. It was randomly divided into sham-operation group and three major groups: subtotal group, subtotal group and total group (each group included: model group, traditional Chinese medicine tea group). There were 12 rats in each of 13 groups of rats in the acupoint injection group and the Chinese herbal tea point injection group. The models were made by three castration methods: total ovariectomies and ovariectomies. The left ovary was excised only about 50 times. Subtotal excision: the right ovary was excised completely, the left ovary was removed only about 80%; Total resection: bilateral ovariectomies. After the model, the traditional Chinese medicine tea group drank freely for 8 weeks. The acupoint injection group selected Guan Yuan and returned (Shuangli, Ganshu (Shuangyou, Shenshu)). Point injection of Danhong injection 0.02mL / d once a week, a total of 8 times. After the end of the last treatment, the serum estradiol estradiol E2 was detected by ELISA method in each group. Follicle stimulating hormone (FSH) and the level of SIRT1 protein in heart tissue; Results: compared with the sham-operated group, the serum E2 level and the SIRT1 protein level in the heart tissue of the model group were significantly lower than those of the sham operation group. The level of serum FSH was significantly increased, and the difference was statistically significant (P 0.05). Compared with the model group, the levels of serum E2 and SIRT1 protein in heart tissue of each treatment group were significantly increased, and the serum FSH levels were significantly decreased. The differences were statistically significant (P 0.05). Among them, the level of E _ 2 and the average value of SIRT1 protein were the highest in the injection group. The average value of FSH water was the lowest. The results of morphological observation of rat endometrium showed that each treatment group could make endometrial epithelium thicker, make interstitial dense, increase glands and enrich blood vessels. Conclusion: in this experiment, we used three castration methods: secondary, subtotal and total resection to make models, and successfully duplicated the model of climacteric rats. And simulated the process of human menopausal ovarian decline. 2) Acupoint injection and traditional Chinese medicine tea can delay the aging process of ovariectomized rats. The mechanism may be that acupoint injection and Chinese herbal tea can increase the level of serum E2 and SIRT1 protein in cardiac tissue of ovariectomized rats. The results showed that the early combined intervention was more effective in the process of ovarian decline in rats.
