七及肠宁灌肠液治疗大鼠溃疡性结肠炎的实验研究

【摘要】:目的:从结肠粘膜屏障修复角度探讨七及肠宁灌肠液治疗大鼠溃疡性结肠炎的疗效。材料与方法:本实验采用2,4,6-三硝基苯磺酸和乙醇混合来创造试验模型,配好的溶液对大鼠进行灌肠,组建溃疡性结肠炎的实验模型。通过记录各组大鼠身体体重的变化、大鼠行为学状态、以及大鼠的粪便情况,椎脱臼处死大鼠。取实验模型大鼠的结肠组织8cm,结肠组织匀浆,测定结肠中EGF、BFGF、CD3和CD8的含量。通过制作组织切片来观察分析;进而探索七及肠宁灌肠液对溃疡性结肠炎大鼠肠粘膜屏障的修护作用。结果:1.通过实验发现X组动物整体状态好转,病变部位治愈恢复相对好,并且直接表现在大鼠的体重增加上。2.模型组CD3和CD8细胞群明显降低,七及肠宁灌肠液高剂量组能显著改善这种情况。3.实验群病变模型创造成功后,模型组EGF和BFGF的升高比其他组明显。结论:1.七及肠宁灌肠液可以改善溃疡性结肠炎模型大鼠脓血便症状,对其肠粘膜屏障有一定修复作用。2.七及肠宁灌肠液治疗溃疡性结肠炎的机理可能是通过下调CD3和CD8的含量,以纠正异常的免疫功能,从而达到消除炎症,修复组织的作用。3.三硝基苯磺酸加乙醇灌肠法造模比较理想,既符合溃疡性结肠炎发生的免疫学机制,又符合肠道炎症发作期的特点,适合于观察药物疗效比较和作用机理探讨研究之用。
[Abstract]:Objective: to investigate the therapeutic effect of Qihehe Changning enema on ulcerative colitis in rats from the point of view of repairing colonic mucosal barrier. Materials and methods: the experimental model was created by mixing 2o 4N 6-trinitrobenzene sulfonic acid with ethanol. The rat model of ulcerative colitis was established by enema with a mixed solution. The changes of body weight, the behavioral state of rats and the feces of rats were recorded by recording the changes of body weight, the behavior of rats, and the feces of rats in each group. The rats were killed by dislocation of vertebrae. The colonic tissue was taken from the experimental model rats at 8 cm and the colonic tissue homogenate was used to determine the contents of CD3 and CD8 in the colon. The contents of CD3 and CD8 were observed and analyzed by making tissue sections. To explore the repairing effect of Qi and Changning enema liquid on intestinal mucosal barrier in rats with ulcerative colitis. Results: 1. It was found that the whole state of the animals in group X was improved and the lesion site was cured and recovered relatively well. The results showed directly in the weight gain of rats. 2. The CD3 and CD8 cell populations in the model group were significantly decreased, and those in the high dose group of 7 and Changning enema solution could significantly improve this situation. 3. After the establishment of the experimental group pathological model, the experimental group lesion model was established successfully. Conclusion the increase of EGF and BFGF in the model group is significantly higher than that in the other groups. Conclusion: 1. 7 and Changning enema can improve the symptoms of pus and stool in rats with ulcerative colitis. The mechanism of repairing intestinal mucosal barrier. 2.7and Changning enema solution in the treatment of ulcerative colitis may be that the abnormal immune function can be corrected by down-regulating the contents of CD3 and CD8 in order to eliminate inflammation. The effect of repairing tissue .3.The method of enema with trinitrobenzene sulfonic acid and ethanol is ideal, which not only accords with the immunological mechanism of ulcerative colitis, but also accords with the characteristics of intestinal inflammatory attack. It is suitable for observing the comparison of drug curative effect and exploring the mechanism of action.
