北五味子多糖对脂多糖诱导中性粒细胞炎症的抑制作用

【摘要】:目的:探讨满药北五味子多糖(NSCP)对脂多糖(LPS)激活的离体培养人中性粒细胞的影响,阐明其抗炎作用机制。方法:利用LPS处理人中性粒细胞建立炎症细胞模型,实验分为对照组、LPS组(1.0mg·L~(-1))及NSCP组(1.25、2.50和5.00g·L~(-1)),NSCP组先加入LPS刺激60min,再加入不同浓度NSCP。应用酶联免疫吸附法(ELISA)检测中性粒细胞中肿瘤坏死因子α(TNF-α)水平,采用流式细胞术检测中性粒细胞凋亡率。结果:与对照组比较,LPS组TNF-α水平升高(P0.05);与LPS组比较,NSCP组TNF-α水平降低(P0.05)。LPS组中性粒细胞凋亡百分率较对照组降低(P0.05);NSCP组中性粒细胞凋亡百分率随时间及剂量增加而升高,其中5.00g·L~(-1) NSCP作用16h促进其凋亡作用最佳,与LPS组比较凋亡百分率明显升高(P0.05)。结论:NSCP可通过抑制LPS诱导TNF-α分泌及中性粒细胞凋亡延迟而发挥抗炎作用。
[Abstract]:Aim: to investigate the effect of NSCP on cultured human neutrophils activated by lipopolysaccharide (LPS), and to elucidate its anti-inflammatory mechanism. Methods: LPS was used to treat human neutrophil to establish inflammatory cell model. The model was divided into two groups: control group (LPS group) and NSCP group (1.25g / L ~ 2.50 and 5.00 g / L ~ (-1)) respectively. The cells were stimulated with LPS for 60 min and then added with different concentrations of LPS. The level of tumor necrosis factor- 伪 (TNF- 伪) in neutrophils was detected by Elisa and the apoptosis rate of neutrophils was detected by flow cytometry. Results: compared with the control group, the level of TNF- 伪 increased in LPS group and decreased in LPS group. The percentage of neutrophil apoptosis in NSCP group was higher than that in control group. Among them, 5.00g / L NSCP had the best effect on promoting apoptosis for 16 h, and the percentage of apoptosis in LPS group was significantly higher than that in LPS group (P 0.05). ConclusionTwo-NSCP can inhibit the secretion of TNF- 伪 and delay the apoptosis of neutrophils by inhibiting the secretion of TNF- 伪 by LPS.
