针刺捻转泻法对应激性高血压前期大鼠下丘脑基因表达谱的影响

【摘要】:目的:探讨针刺捻转泻法对应激性高血压前期(SI-PHT)大鼠的作用机制。方法:Wistar大鼠随机分为空白组(12只)、模型组(12只)和捻转泻法组(10只)。以足底电击结合噪声刺激方法复制SI-PHT模型。造模期间,每日对捻转泻法组大鼠双侧"太冲""曲池"行针刺捻转泻法操作。实验前1天,第3、5、7、9、11天测量各组大鼠收缩压,并观察行为学变化;以基因芯片技术检测并分析各组下丘脑基因表达谱。结果:与空白组相比,第3~11天模型组和捻转泻法组大鼠收缩压均明显升高(P0.01);与模型组相比,第5~11天捻转泻法组大鼠收缩压下降(P0.01);模型组和捻转泻法组大鼠同时伴躁狂等行为学改变,但捻转泻法组较模型组减轻。40条基因在模型组与空白组、捻转泻法组与模型组的比较中表达趋势均相反(P0.05,|FC|≥1.5);其中,Acsm 3、Cgα、CryαB和Tshβ与高血压明显相关(P0.05,|FC|≥1.5);捻转泻法通过激活Autoimmune thyroid disease(AITD)通路发挥降压效应。结论:针刺捻转泻法可引起SI-PHT大鼠下丘脑基因Acsm 3、Cgα、CryαB和Tshβ的激活,出现生物学及分子功能的变化,并通过激活AITD通路发挥降压效应。
[Abstract]:Objective: to investigate the mechanism of acupuncture twisting and reducing method on SI-PHT rats. Methods Wistar rats were randomly divided into blank group (n = 12) and model group (n = 12) and control group (n = 10). SI-PHT model was established by plantar shock combined with noise stimulation. During the modeling period, acupuncture twisting and catharsis were performed on both sides of the rats in the twisting and catharsis group. Systolic blood pressure (SBP) was measured and behavioral changes were observed one day before the experiment, and gene expression profiles of hypothalamus in each group were detected and analyzed by gene chip technique. Results: compared with the blank group, the systolic blood pressure of the rats in the model group and the twirling diarrhea group were significantly increased on the 3rd day and 11th day, and compared with the model group. The rats in the model group and the twisting method group were accompanied with mania and other behavioral changes at the same time, but the rats in the twisting and reducing method group had less .40 genes in the model group and blank group, and the rats in the model group and the twisting and reducing method group had decreased the number of .40 genes in the model group and the blank group.
