板桥党参多糖对LPS诱导的ATDC-5细胞NO、IL-1β及Ⅱ型胶原影响的实验研究

【摘要】:目的:体外培养软骨细胞系ATDC-5细胞,观察板桥党参多糖的干预对ATDC-5细胞活性的影响及NO的释放量、i NOs的活力、IL-1β、II型胶原的表达影响,为丰富益气法治疗OA提供理论依据。方法:采用水提醇沉法提取板桥党参多糖;体外培养ATDC-5细胞,CCK-8法检测板桥党参多糖对ATDC-5细胞活性的影响;将ATDC-5细胞分为5组,分别为空白组、LPS组、板桥党参多糖组(低浓度组、中浓度组、高浓度组)。硝酸还原酶法检测NO的释放量、ELISA法检测IL-1β的表达情况、Western Blot法检测II型胶原的表达情况。结果:1提取板桥党参多糖4.53g;2不同浓度板桥党参多糖作用于ATDC-5细胞,1000μg/ml的多糖溶液能在一定程度抑制ATDC-5细胞的增殖活性,与空白组相比较P0.05,差异有统计学意义;3在LPS的作用下,ATDC-5细胞i NOs的活力、NO的释放量、IL-1β的量均有所增加,与空白组相比较P0.05,差异有统计学意义;而加入板桥党参多糖后,i NOs的活力、NO的释放量、IL-1β的量明显减少;4在LPS作用下,ATDC-5细胞表达的II型胶原会明显减少与空白组相比较P0.05,差异有统计学意义,而板桥党参多糖能对抗LPS抑制ATDC-5细胞合成II型胶原的作用。结论:1 LPS能刺激ATDC-5细胞产生大量的NO;2板桥党参多糖能一定程度抑制LPS诱导ATDC-5细胞NO的释放(通过抑制i NOs活性);3板桥党参多糖能一定程度抑制LPS诱导的ATDC-5细胞IL-1β的表达;4板桥党参多糖能对抗LPS抑制ATDC-5细胞合成II型胶原的作用,使ATDC-5细胞在受到炎性刺激的时候还能表达相对正常的II型胶原。
[Abstract]:Objective: to culture chondrocyte ATDC-5 cells in vitro, observe the effect of Codonopsis pilosula polysaccharide on the activity of ATDC-5 cells and the expression of IL-1 尾 type II collagen in the release of no and the activity of I NOs, and provide a theoretical basis for enriching the method of supplementing qi to treat OA. Methods: the polysaccharides of Codonopsis pilosula were extracted by water extraction and alcohol precipitation, the effect of polysaccharides of Codonopsis pilosula on the activity of ATDC-5 cells was detected by CCK-8 method in vitro cultured ATDC-5 cells, and the ATDC-5 cells were divided into five groups: blank group, low concentration group. Medium concentration group, high concentration group. No release was detected by nitrate reductase method and the expression of IL-1 尾 by Elisa and type II collagen by Western Blot. Results the polysaccharides of Panqiao Codonopsis pilosula with different concentrations (4.53g / g) could inhibit the proliferative activity of ATDC-5 cells in 1 000 渭 g/ml solution of 1 000 渭 g/ml in ATDC-5 cells. Compared with the control group (P0.05), the difference was statistically significant (P 0.05). The activity of NOs in ATDC-5 cells induced by LPS was increased and the amount of IL-1 尾 was increased. The difference was statistically significant compared with the control group (P0.05). However, the activity of NOs and the amount of IL-1 尾 after adding Panqiao Codonopsis pilosula polysaccharides decreased significantly the expression of type II collagen in ATDC-5 cells induced by LPS was significantly decreased compared with the control group (P 0.05), and the difference was statistically significant. The polysaccharide of Codonopsis pilosula could inhibit the synthesis of type II collagen by LPS. Conclusion 1 LPS can stimulate the production of a large amount of No2 Codonopsis pilosula polysaccharide from ATDC-5 cells. It can inhibit the release of no from ATDC-5 cells induced by LPS to some extent (by inhibiting the activity of I NOs), Codonopsis lanceolata polysaccharide can inhibit IL-1 尾 of ATDC-5 cells induced by LPS to some extent. Dangshen polysaccharide could antagonize the effect of LPS on the synthesis of type II collagen in ATDC-5 cells. ATDC-5 cells were also able to express type II collagen in response to inflammatory stimulation.
