电针对去卵巢大鼠骨密度及下丘脑中瘦素与瘦素受体的影响

【摘要】:目的:通过观察电针对去卵巢雌性SD大鼠骨密度及下丘脑中瘦素与瘦素受体含量的影响,探讨针刺治疗绝经后骨质疏松症的可能机制,为针刺治疗绝经后骨质疏松症的临床应用提供实验依据。方法:将40只3月龄健康雌性SD大鼠随机分为4组:假手术组、模型组、药物组、电针组,每组10只。其中假手术组去除卵巢周围脂肪组织,其余三组去除卵巢,手术造模结束后将大鼠适应性喂养5天,待情况稳定后进行干预。电针组予以电针肾俞、足三里、关元、三阴交四穴并灌服生理盐水,药物组给予阿仑膦酸钠并行固定处理,假手术组与模型组行固定处理并同时灌服生理盐水,4组SD大鼠均每天干预1次,10次1个疗程,共治疗9个疗程。疗程结束后剔除大鼠右侧股骨并分离出下丘脑,采用双能X线骨密度仪检测股骨骨密度,RT-PCR法测定下丘脑中LP m RNA、OB-R m RNA的定量。结果:1.大鼠行卵巢摘除后骨密度开始降低,疗程结束后发现模型组、药物组、电针组骨密度明显低于假手术组(P0.05),但电针组、药物组骨密度值高于模型组(P0.05)。2.模型组下丘脑中LP及OB-R的含量高于假手术组(P0.05),电针组和药物组下丘脑中LP及OB-R的含量低于模型组(P0.05)。结论:1.电针肾俞、足三里、关元、三阴交能够减少雌性SD大鼠去卵巢后骨量丢失,能够防治绝经后骨质疏松症。2.电针肾俞、足三里、关元、三阴交能够抑制去卵巢后大鼠下丘脑中LP及OB-R含量的升高,削弱其骨形成中枢抑制作用,此调节方式可能具有减少绝经后骨量丢失的作用。
[Abstract]:Objective: to investigate the possible mechanism of acupuncture in treating postmenopausal osteoporosis by observing the effects of electroacupuncture on bone mineral density (BMD) and the contents of leptin and leptin receptor in hypothalamus of ovariectomized female SD rats. To provide experimental evidence for the clinical application of acupuncture in the treatment of postmenopausal osteoporosis. Methods: forty healthy female SD rats aged 3 months were randomly divided into 4 groups: sham operation group, model group, drug group, electroacupuncture group, 10 rats in each group. The sham-operation group removed the periovarian adipose tissue and the other three groups removed the ovary. The rats were fed adaptively for 5 days after the operation. The electroacupuncture group was given electroacupuncture of Shenshu, Zusanli, Guan Yuan, Sanyinjiao four points and normal saline, while the drug group was treated with alendronate and fixed simultaneously. The sham operation group and the model group were treated with fixed treatment and were treated with saline at the same time. The SD rats in the 4 groups were treated with 10 times a course of treatment once a day for 9 courses of treatment. After the course of treatment, the right femur was removed and the hypothalamus was isolated. The bone mineral density (BMD) of femur was measured by dual-energy X-ray absorptiometry. The OB-R m RNA in the hypothalamus was measured by RT-PCR. The result is 1: 1. The BMD of the model group, drug group and electroacupuncture group was significantly lower than that of the sham operation group, but the BMD in the electroacupuncture group was higher than that in the model group. The content of LP and OB-R in hypothalamus of model group was higher than that of sham operation group, and the content of LP and OB-R in hypothalamus of electroacupuncture group and drug group was lower than that of model group. Conclusion 1. Electroacupuncture of Shenshu, Zusanli, Guan Yuan and Sanyinjiao can reduce bone loss after ovariectomy in female SD rats, and can prevent and treat postmenopausal osteoporosis. Electroacupuncture of Shenshu, Zusanli, Guan Yuan and Sanyinjiao could inhibit the increase of LP and OB-R content in hypothalamus of ovariectomized rats, and weaken the central inhibition of bone formation.
