丹贝益肺方对肺纤维化大鼠肺组织MMP-9和Egr-1表达影响的研究

【摘要】:目的:通过观察丹贝益肺方作用于肺纤维化大鼠后,其肺组织中MMP-9和Egr-1表达的变化,及大鼠肺纤维化的改善情况,以验证丹贝益肺方的疗效,并探讨其作用机理。方法:取90只健康大鼠(Wistar),在清洁级实验室正常饲养一周。随机平均分成5组,每组雌雄等量。其中1组用生理盐水造模,标记为空白组。其余4组均用博莱霉素造模,分别标记为:模型组,泼尼松(阳性对照)组,中药(丹贝益肺)高、低剂量组。造模成功后,分别给与0.9%生理盐水,泼尼松水溶液(浓度0.5mg/ml),丹贝益肺高剂量水煎液(浓度1.2g/ml),丹贝益肺低剂量水煎液(浓度0.3g/ml),均按体重比例(10ml/kg)灌胃。实验期间观察大鼠一般状态,并在第7天、14天、28天时,各组随机处死6只大鼠并取材,测定大鼠肺系数,观察肺病理组织形态学变化,同时用相关分级评分标准对肺组织肺泡炎及纤维化程度进行评分,并用免疫组织化学法检测大鼠肺组织中MMP-9和Egr-1的表达水平。结果:中药各剂量组及泼尼松组大鼠肺系数与模型组同期组间比较,均降低(P0.05),对照相应病理组织切片观察结果,提示泼尼松、丹贝益肺方高低计量均可减轻大鼠肺泡炎程度、减缓纤维化进展。泼尼松组、中药各剂量组大鼠的肺泡炎积分及纤维化积分低于同期模型组(P0.05),进一步验证泼尼松和中药高低剂量对大鼠肺泡炎及肺纤维化均有治疗作用。与同期空白组相比,其余各组大鼠肺组织中MMP-9和Egr-1表达均增高(P0.05),提示肺纤维化的形成与MMP-9及Egr-1有关;同期泼尼松组、中药各剂量组肺组织中MMP-9和Egr-1表达均低于模型组,具有统计学差异性(P0.05),说明丹贝益肺方对肺纤维化的治疗机制可能与降低MMP-9及Egr-1的表达具有相关性。同期大鼠一般状态,泼尼松组及中药各剂量组均优于模型组,说明泼尼松及丹贝益肺方可使肺纤维化大鼠的症状得到改善。结论:丹贝益肺方可改善肺纤维化大鼠症状表现,降低大鼠肺系数,减轻其肺泡炎及肺纤维化程度,减低肺组织中MMP-9和Egr-1的表达,且该表达的减少可能是丹贝益肺对肺纤维化的治疗机制之一。
[Abstract]:Aim: to observe the changes of MMP-9 and Egr-1 expression in lung tissue and the improvement of pulmonary fibrosis in rats after the action of Danbei Yifei recipe on pulmonary fibrosis in order to verify the curative effect of Danbei Yifei recipe and to explore its mechanism. Methods: 90 healthy rats were collected and fed in clean laboratory for one week. They were randomly divided into 5 groups with equal amount of both sexes in each group. The model was made with normal saline in group 1 and marked as blank group. The other four groups were made with bleomycin, which were labeled as model group, prednisone (positive control) group, high and low dose group of traditional Chinese medicine (Danbei Yifei). After the establishment of the model, 0.9% normal saline was given, prednisone solution (0.5 mg / ml), high dose Danbei Yifei decoction (1.2 g / ml) and low dose (0.3 g / ml 路ml ~ (-1) Danbei Yifei decoction (0.3 g / ml ~ (-1) 路kg ~ (-1) respectively. During the experiment, the general state of the rats was observed, and on the 7th day, the 14th day and the 28th day, 6 rats were killed randomly in each group, and the lung coefficient was measured, and the pathological changes of the lung were observed. At the same time, the degree of pulmonary alveolitis and fibrosis was evaluated by the correlation grading standard, and the expression of MMP-9 and Egr-1 in the lung tissue of rats was detected by immunohistochemical method. Results: compared with the model group, the lung coefficient of each dosage group and prednisone group decreased P0.05, and compared with the corresponding pathological tissue section observation, it suggested that prednisone and Danbei Yifei prescription could reduce the degree of alveolitis in rats. Slow the progression of fibrosis. The alveolitis score and fibrosis score of rats in prednisone group and traditional Chinese medicine group were lower than those in the model group (P 0.05). It was further proved that prednisone and high and low doses of traditional Chinese medicine had therapeutic effects on alveolitis and pulmonary fibrosis in rats. Compared with the blank group, the expression of MMP-9 and Egr-1 in the lung tissues of the other groups were all higher than that of the control group, indicating that the formation of pulmonary fibrosis was related to MMP-9 and Egr-1, and the expression of MMP-9 and Egr-1 in the lung tissues of the prednisone group and the traditional Chinese medicine group were all lower than those of the model group. There was statistical difference (P 0.05), which indicated that the therapeutic mechanism of Danbei Yifei recipe on pulmonary fibrosis might be related to the decrease of MMP-9 and Egr-1 expression. At the same time, the general state of rats, prednisone group and traditional Chinese medicine groups were better than the model group, indicating that prednisone and Danbei Yifei Fang could improve the symptoms of pulmonary fibrosis rats. Conclusion: danbei Yifei decoction can improve the symptoms of rats with pulmonary fibrosis, reduce the lung coefficient, reduce the degree of alveolitis and pulmonary fibrosis, and reduce the expression of MMP-9 and Egr-1 in lung tissue. The decrease of this expression may be one of the therapeutic mechanisms of lung fibrosis.
