基于“共同土壤”学说建立2型糖尿病合并冠心病寒凝血瘀证大鼠模型的研究

【摘要】:实验目的:基于“共同土壤”学说探索建立2型糖尿病合并冠心病大鼠动物模型,并根据“寒独留则血凝泣,凝则脉不通”的中医理论在前者基础上进一步建立2型糖尿病合并冠心病寒凝血瘀证大鼠模型。实验方法:运用高脂高糖饲料喂养小剂量腹腔注射链脲佐菌素(STZ)以及维生素D3腹腔注射并加上冰浴方法进行造模。应用量化评分表对造模期间大鼠的症状和体征进行评分,并在造模的第30、60、90天以及注射STZ后行眼眶采血,用全自动生化仪以及血脂检测试剂盒测定大鼠血脂,用血糖检测仪检测大鼠空腹血糖,用全自动血流变检测血液流变学改变以及用ELESA法检测大鼠血清胰岛素、前列腺素和血栓素的变化,用心电图机记录大鼠心电图Ⅱ导联ST段的改变,并于造模结束后对大鼠的心脏与胰腺进行HE染色以观察其组织形态的改变。实验结果:(1)模型组大鼠在症状与体征方面出现毛发竖立、枯萎,畏寒喜暖,喜爱扎堆,行动迟钝,活动减少,饮水以及摄食量均减少,体重减轻,大便稀软,耳缘、爪甲紫暗,舌质暗红有瘀斑,舌下络脉曲张等寒凝血瘀证症状。(2)模型组大鼠血脂、血糖、全血黏度均增高,与空白组对照有显著性差异(P0.05,P0.01)。(3)模型组大鼠心电图II导联ST段有显著下移现象,与空白组对照有显著性差异(P0.01)(4)模型组大鼠血清胰岛素、前列腺素、血栓素均出现紊乱,与空白组对照有显著性差异(P0.05,P0.01)。(5)模型组大鼠心脏组织出现冠状动脉硬化、心肌细胞水肿、坏死,胰腺组织出现胰岛细胞萎缩破坏等病理学改变。实验结论:运用高脂高糖饲料喂养小剂量腹腔注射链脲佐菌素(STZ)以及维生素D3灌胃并加上冰浴方法建立的Ⅱ型糖尿病合并冠心病寒凝血瘀证与中医证侯特点相符合,具有一定的理论和实践意义。
[Abstract]:Objective: to establish an animal model of type 2 diabetes mellitus with coronary heart disease (CHD) based on the theory of "common soil". On the basis of the former, the model of type 2 diabetes mellitus with cold coagulation and blood stasis syndrome of coronary heart disease was established. Methods: low dose of STZ and vitamin D3 were fed with high fat and high sugar diet, and the model was made by ice bath. The symptoms and signs of the rats during model making were scored by using the quantitative scoring table. The blood samples were collected from the orbit on the 30th day of model making and after STZ injection. The blood lipid of the rats was measured by automatic biochemical instrument and blood lipid test kit. Fasting blood glucose was detected by blood glucose detector, hemorheology was detected by automatic hemorheology and serum insulin, prostaglandin and thromboxane were detected by ELESA method.
