异常黑胆质成熟剂对抑郁症大鼠模型支配器官组织形态学的影响

【摘要】:目的观察异常黑胆质成熟剂(ASMq)对抑郁症大鼠模型“支配器官”(脑、心、肝)病理变化和超微结构的影响,从组织形态学角度探讨ASMq抗抑郁的可能作用机制。方法将雄性抑郁症模型大鼠60只,机分为5组:异常黑胆质成熟剂高、中、低剂量组(ASMq给药剂量为6.0、3.0、1.5 g/kg),阳性对照组(氟西汀,3.5g/kg),抑郁症模型对照组,每组12只,另设正常对照组(12只)。ASMq各剂量组和阳性对照组连续灌胃给药21d,1次/天,末次给药1h后颈椎脱臼处死动物取出支配器官固定,HE染色,采用光学显微镜观察“支配器官”病理变化,铅铀染色,制作常规电镜标本,扫描电镜观察支配器官超微结构改变。结果与模型组相比,ASMq高剂量组“支配器官”细胞修复性增强,而损伤性组织病理学改变显著减轻,ASMq低、中剂量组“支配器官”损伤性组织病理变化无显著差异。抑郁症模型组“支配器官”超微结构中除了脑组织神经元细胞外,其余组织细胞有不同程度的细胞水肿、基质密度明显降低,细胞变质等超微结构的损伤,但ASMq干预后,脑神经胶质细胞水肿明显降低、细胞基质密度、细胞器稀疏明显恢复,神经细胞突触部脑水肿明显的降低、可见轻微空泡样变;心肌细胞水肿明显消退、心肌细胞基质密度恢复明显,未见肌原纤维局灶性肌丝溶解,而心肌细胞内线粒体水肿明显消退,未见局部有异常收缩带;肝细胞水肿明显消退,基质密度明显恢复、细胞内可见轻微空泡、肝细胞内可见脂滴减小、肝细胞间隙增宽恢复明显、毛细胆管未扩张,肝细胞间未见胶原纤维增生。结论异常黑胆质成熟剂能够改善抑郁症大鼠模型动物“支配器官”病理变化和超微结构的改变。
[Abstract]:Objective to observe the effects of abnomal black bile maturation (ASMQ) on the pathological changes and ultrastructure of brain, heart and liver in a rat model of depression, and to explore the possible mechanism of antidepressant effect of ASMq in terms of histomorphology. Methods Sixty male depression model rats were divided into 5 groups. The dosage of ASMq was 6.0 ~ 3.0 ~ 1.5 g 路kg ~ (-1) 路kg ~ (-1) in the medium and low dose groups, 12 rats in the depression model control group were treated with fluoxetine 3.5 g / kg 路kg ~ (-1), depression model control group (n = 12). In addition, 12 rats in the normal control group and the positive control group were given intragastric administration of the drug for 21 days. The animals were killed for 1 hour after the last administration of the drug. The animals were killed to remove the innervation organ for HE staining, and the pathological changes of the "innervation organ" were observed by optical microscope. The ultrastructural changes of dominant organs were observed by scanning electron microscope (SEM). Results compared with the model group, the repair ability of the "innervation organ" cells in the high dose group of ASMq was enhanced, while the pathological changes of the injured tissue were significantly reduced, but there was no significant difference in the pathological changes of the damaged tissue of the "innervated organ" in the middle dose group. In the model group of depression, the ultrastructure of "innervation organ" was damaged by cell edema, matrix density, cell degeneration and other ultrastructural damage except neurons in brain tissue, but after ASMq intervention. The edema of glial cells, the density of cell matrix, the thinning of organelle, the brain edema in synaptic part of nerve cells, the slight vacuolation, the edema of cardiac myocytes, and the edema of cardiac myocytes disappeared obviously. The matrix density of cardiomyocytes recovered obviously, and no focal myofilament lysis was found in myofibrils, while mitochondria edema disappeared obviously and there was no abnormal contractile zone in myocardial cells, the edema of hepatocytes subsided obviously and the matrix density recovered obviously. There were slight vacuoles in the cells, lipid droplets decreased in the hepatocytes, the gap widened obviously, the capillary bile ducts did not dilate, and the collagen fibers were not proliferated among the hepatocytes. Conclusion abnormal melanoglycoid maturation agent can improve the pathological changes and ultrastructural changes of "innervation organ" in rats with depression.
