黄葵素对糖尿病肾病大鼠中HB-EGF表达的影响

【摘要】:目的:糖尿病肾病(Diabetic Nephropathy, DN)是糖尿病患者微血管并发症之一,发病机制及相关治疗是世界医学面临的一大难题,研究DN的道路任重道远。本实验通过研究肝素结合表皮生长因子样生长因子(heparin-binding epidermal growth factor-like growth factor, HB-EGF)在DN模型大鼠血清和肾脏组织中的表达以及黄葵素的干预作用,对黄葵素治疗DN的可能机制作一个初步的探讨。方法:首先从60只SPF级雄性的SD大鼠中随机抽取10只,设为空白组,其余50只则为造模组。空白组的大鼠给予普通饲料饲养,造模组所有大鼠均采用高脂饮食联合小剂量STZ腹腔注射的方法建立DN大鼠模型。再将DN造模成功的大鼠随机分成模型组、阳性对照组以及黄葵素高、中、低剂量组,每组10只。阳性对照组的大鼠予厄贝沙坦溶液灌胃给药,黄葵素高、中、低剂量组的大鼠分别给予高、中、低浓度黄葵素混悬液灌胃给药;空白组、模型组的大鼠都采用双蒸水灌胃。给药12周后收集标本,静脉血用于测血糖;大鼠尿液用于U-mAlb/Cr的测定;血清用ELASA法行HB-EGF定量检测;肾脏组织运用荧光定量RT-PCR法检测HB-EGF总mRNA的表达。结果:与空白组相比,模型组、阳性对照组以及黄葵素高、中、低剂量组大鼠的血糖、尿微量白蛋白/肌酐、血清HB-EGF浓度水平、肾组织中HB-EGF分子mRNA的表达量均明显升高,且具有统计学差异。阳性对照组、黄葵素高剂量组大鼠血糖、尿微量白蛋白/肌酐、血清HB-EGF浓度水平、肾组织中HB-EGF分子mRNA的表达量均低于模型组大鼠,差异有统计学意义。黄葵素中、低剂量组大鼠血糖、尿微量白蛋白/肌酐、血清HB-EGF浓度水平、肾组织中HB-EGF分子mRNA的表达量与模型组大鼠无显著差异。结论:DN模型大鼠存在肾脏损害的同时伴有HB-EGF表达的增多;运用黄葵素对这类大鼠进行干预后,各组大鼠血糖、尿蛋白均得到一定程度的控制,同时大鼠血清和肾脏组织中]HB-EGF的表达也相应的减少,并且存在一定的量效关系,因此,黄葵素治疗DN可能与通过抑制HB-EGF的表达有关。
[Abstract]:Objective: diabetic nephropathy (DN) is one of the microvascular complications in diabetic patients. The purpose of this study was to investigate the expression of heparin-binding epidermal growth factor-like growth factor (HB-EGF) in serum and kidney of DN model rats and the intervention effect of flavitin on the expression of heparin-like growth factor-like growth factor (HB-EGF) in serum and kidney of DN rats. To explore the possible mechanism of sunflower in the treatment of DN. Methods: ten SPF male SD rats were randomly selected as blank group and the other 50 rats as model group. The rats in the blank group were fed with common diet and all the rats in the model group were fed with high fat diet combined with small dose STZ intraperitoneal injection to establish DN rat model. The rats were randomly divided into the model group, the positive control group and the high, middle and low dosage groups with 10 rats in each group. The rats in the positive control group were given irbesartan solution by gavage, and the rats in the high, middle and low dose groups were given the high, middle and low concentration of flavitin suspension respectively, while in the blank group, the rats in the model group were fed with double-steamed water respectively. After 12 weeks of administration, blood samples were collected and blood was used to measure blood glucose; rat urine was used to determine U-mAlb/Cr; serum ELASA was used for quantitative detection of HB-EGF; and renal tissue was used to detect the expression of total mRNA of HB-EGF by fluorescence quantitative RT-PCR method. Results: compared with the blank group, the levels of blood glucose, urinary microalbumin / creatinine, serum HB-EGF concentration, and the expression of HB-EGF molecule mRNA in kidney were significantly increased in the model group, positive control group and high, middle and low dose groups. And there is statistical difference. In the positive control group, the levels of blood glucose, urinary albumin / creatinine, serum HB-EGF concentration, and the expression of HB-EGF molecule mRNA in renal tissue were significantly lower in the high-dose group than in the model group. There was no significant difference in the levels of blood glucose, urinary albumin / creatinine, serum HB-EGF concentration and the expression of HB-EGF molecule mRNA between the low dose group and the model group. Conclusion there is renal damage and increase of HB-EGF expression in the rats with 1: DN model, and the blood glucose and urine protein in each group are controlled to some extent after the intervention of horseradine on these rats, and the results are as follows: (1) the blood glucose and urine protein of the rats in each group were controlled to a certain extent. At the same time, the expression of HB-EGF in serum and kidney of rats was also decreased, and there was a dose-effect relationship. Therefore, the treatment of DN with flavidin may be related to the inhibition of HB-EGF expression.
