壮药龙船花和露兜树的质量标准研究

【摘要】:目的:对壮药龙船花(Ixora chinensis Lam.)和露兜树(Pandanus tectorius Soland.)的质量标准进行研究,建立和评价龙船花、露兜树药材质量标准的方法,并制订壮药龙船花和露兜树的质量标准,为壮药龙船花和露兜树的质量控制提供科学的依据。方法:(1)从龙船花和露兜树的根或茎横切面、药材粉末特征、植物形态、药材性状鉴别等多方面进行生药学研究;(2)通过对药材的水分测定,总灰分测定、酸不溶性灰分测定及浸出物测定对龙船花和露兜树药材进行检查项研究;(3)通过化学成分预试验,对龙船花和露兜树进行了化学成分初步研究;(4)通过薄层鉴别法对龙船花和露兜树药材进行定性鉴别;(5)通过HPLC对龙船花中东莨菪内酯的含量进行测定;(6)参照2015年版《中华人民共和国药典》制订了壮药龙船花和露兜树的药材质量标准(草案)。结果:(1)生药学研究龙船花:茎呈圆柱形,黄褐色,直径5~8mm,断面皮部薄,黄褐色或黑褐色,木部灰白色,叶对生,花序顶生多花。木栓层有数列厚角组织细胞,形成层明显,木质部相对较宽并具有大量木纤维,中央髓部宽广,有很多大型石细胞群。气孔为平轴式,复粒淀粉粒由2~3个单粒淀粉粒组成,草酸钙簇晶和棕色体常见,螺纹导管较多,孔沟明显。露兜树:根呈圆柱形,棕黄色,直径3~6mm,皮部皱缩成棱状突起,叶缘和背面中脉均有粗壮的锐刺,聚花果大,核果束倒圆锥形,质坚硬。皮部宽广,有大量纤维束和大型黏液细胞,内皮层明显,中柱维管束成辐射状。淀粉粒类圆形,复粒淀粉粒,草酸钙针晶较多,常见晶鞘纤维,导管主为螺纹、梯纹、具缘纹孔导管,气孔平轴式。(2)检查项龙船花:通过对壮药龙船花10个不同批号的样品进行研究,10批样品水分含量拟订为不得过12.0%,总灰分拟定为不得过6.0%,酸不溶性灰分拟定为不得过1.5%,浸出物含量拟订为不少于13.0%。露兜树:通过对壮药露兜树10个不同批号的样品进行研究,10批样品水分含量拟订为不得过9.6%,总灰分拟定为不得过8.0%,酸不溶性灰分拟定为不得过2.0%,浸出物含量拟订为不少于12.5%。(3)化学成分预试验龙船花药材中可能含有糖、多糖、苷类、皂苷、鞣质、黄酮类、植物甾醇、三萜类及香豆素类等成分。露兜树药材中可能含有糖、多糖、苷类、有机酸、黄酮类、酚类、香豆素、内酯等成分。(4)药材供试品薄层色谱中,在与对照药材色谱相应的位置上,显相同颜色的斑点,重现性较好。(5)10批样品中含量最高的是LCH-2=0.06103mg/g;含量最低的是LCH-3=0.04843mg/g,10批样品平均含量(0.055415mg/g),降低20%计,拟定壮药龙船花东莨菪内酯含量限度不低于0.044332mg/g(按干燥品计算,不得少于0.0044332%)。结论:本文对龙船花和露兜树的根或茎横切面、药材粉末特征、植物形态、药材性状鉴别的生药学研究具有一定意义。通过薄层鉴定对龙船花和露兜树药材进行了定性鉴别。检查项研究和含量测定结果为龙船花和露兜树药材质量控制提供了评价指标。参照2015年版《中华人民共和国药典》制订了龙船花和露兜树的药材质量标准草案,该标准具有科学性、可行性、实用性等特点,为龙船花和露兜树药材质量控制提供参考依据。
[Abstract]:Objective: To study the quality standard of Ixora chinensis Lam. and Pandanus tectorius Soland., to establish and evaluate the quality standard of the Dragon Boat flower and the crude drug, and to establish the quality standard of the Dragon Boat flower and the drive tree of the strong medicine, and provide the scientific basis for the quality control of the Dragon Boat flower and the radix Acacus. Methods: (1) the study of Pharmacognosy from the root or stem of the dragon boat and the root or stem of the tree, the characteristics of the medicinal powder, the plant morphology, the identification of the medicinal properties, and so on. (2) the determination of the moisture of the medicinal materials, the determination of the total ash, the determination of acid insoluble ash and the determination of the extracts of the extract on the medicinal herbs of the dragon boat and the radix acacinus; (3) through chemical formation. Preliminary study on the chemical composition of dragon boat flower and Lusa tree; (4) qualitative identification of dragon boat flower and Radix acurus by TLC identification; (5) the content of scopolamine in Dragon Boat flower was measured by HPLC; (6) according to the 2015 edition of People's Republic of China Pharmacopoeia, the drug of Dragon Boat flower and luschini tree was formulated. Material quality standard (Draft). Results: (1) pharmacognosy study Dragon Boat flower: the stem is cylindrical, yellow brown, 5~8mm diameter, the skin of the section is thin, yellow brown or black brown, the wood is gray, the leaf is green and the inflorescence is most flower. The cork layer has a number of thick angle tissue cells, the formation layer is obvious, the xylem is relatively wide and has a large number of wood fiber, the central pith is broad, the central pith is broad, central pulp. There are a lot of large stone cell groups. The stoma is a horizontal axis. The starch granules are composed of 2~3 single starch granules. The calcium oxalate clusters and brown bodies are common, the threaded ducts are more and the groove is obvious. The roots show a cylindrical, brown yellow, 3~6mm diameter, and the skin shrinks in the prismatic protuberance. The leaf margin and the back of the middle vein have strong sharp spines, large PPS, and Ribas. The beam is conical and hard. The skin is broad and broad, with a large number of fiber bundles and large mucous cells. The endothelial layer is obvious. The vascular bundle of the middle column is radiated. The starch granules are round, the starch granules, the calcium oxalate needle are more, the common crystal sheath fiber, the main thread of the duct, the ladder and the stomata flat axis. (2) check the Dragon Boat flower: through the Zhuang Medicine 10 samples of different batch numbers of the dragon boat were studied. The water content of the 10 batches was drawn up to no more than 12%, the total ash was drawn up to not over 6%, the acid insoluble ash was drawn up to no more than 1.5%, the content of the extract was not less than the 13.0%. tree: the samples of 10 different batch numbers of the strong drug Bib tree were studied and the moisture of the 10 batch of samples was found. The content is not over 9.6%, the total ash can not be over 8%, acid insoluble ash is drawn up to not over 2%, the content of extract is not less than 12.5%. (3) chemical composition pretest Dragon Boat flower medicinal herbs may contain sugar, polysaccharide, glycosides, tannins, Huang Tong, phytosterols, three terpenoids and coumarins. It may contain sugar, polysaccharides, glycosides, organic acids, flavonoids, phenols, coumarins, lactone and other components. (4) in the thin layer chromatography of drug supply samples, the same color spots and reproducibility are found in the corresponding location of the drug chromatography. (5) the highest content of the 10 batches is LCH-2=0.06103mg/g; the lowest content is LCH-3=0.04843mg/g, 10 batches. The average content of the sample (0.055415mg/g) was reduced by 20%, and the limit of the content of scopolamine was not less than 0.044332mg/g (no less than 0.0044332%). Conclusion: the Pharmacognosy study of the root or stem transverse section of the Dragon Boat flower and the rhizome of the rhizome of the dragon boat, the characteristics of medicinal powder, plant morphology and the identification of medicinal properties The qualitative identification was carried out by TLC identification. The results of examination and content determination provided the evaluation index for the quality control of the Dragon Boat flower and the Chinese medicinal herb. According to the 2015 edition of the People's Republic of China Pharmacopoeia, the draft quality standard of the Dragon Boat flower and the wild tree was formulated. The characteristics of learning, feasibility and practicability provide reference for quality control of medicinal materials of dragon boat flower and wild flower tree.
