虫草保肾颗粒对慢肾衰大鼠肾功能及高脂高凝状态的影响

【摘要】:目的:观察虫草保肾颗粒对慢性肾衰竭大鼠肾功能、血脂和同型半胱氨酸水平的影响,探讨其能否通过调控血脂和同型半胱氨酸水平而改善大鼠高脂高凝状态。方法:将78只雄性SD大鼠称重后,随机分为对照组(26只),模型组(26只),虫草保肾颗粒组(26只)。三组均给予普通饲料喂养,模型组及虫草组均采用5/6肾切除术制作CRF模型。给药方案:造模后,各组开始灌胃给药,每日一次,持续12周,虫草保肾颗粒组给予虫草保肾颗粒水煎剂灌胃,对照组和模型组给予等量蒸馏水灌胃。分别于造模后0、4、8、12周观察大鼠一般状态,并进行24小时尿蛋白定量及血清肌酐(Scr)、尿素氮(BUN)、胆固醇(CHOL)、甘油三酯(TG)、低密度脂蛋白(LDL-C)和同型半胱氨酸(HCy)水平测定,同一时间采样点每组大鼠处死6只,并采用HE染色法观察肾组织形态学改变。结果:1.虫草保肾颗粒能有效改善大鼠一般状态;2.虫草保肾颗粒可以降低CRF大鼠血清Scr、BUN,降低蛋白尿水平,与模型组比较有显著差异(P0.01,P0.05);3.虫草保肾颗粒可以降低CRF大鼠血清CHOL、TG、LDL-C和Hcy水平,与模型组比较有显著差异(P0.01,P0.05);4.经虫草保肾颗粒治疗后,肾小球硬化程度、间质纤维化程度、肾小管炎性细胞浸润程度均有减轻。结论:1.虫草保肾颗粒能够改善CRF大鼠一般状态,延缓病情进展;2.虫草保肾颗粒可通过降低CRF大鼠血清Scr、BUN水平,减轻蛋白尿,从后延缓病情进展;3.虫草保肾颗粒可以通过降低血清CHOL、TG、LDL-C和Hcy水平,改善肾衰后期高脂高凝状态;4.虫草保肾颗粒可以减轻5/6肾切除大鼠肾小球硬化和间质纤维化程度。
[Abstract]:Aim: to observe the effects of Cordyceps baoshen granule on renal function, blood lipid and homocysteine levels in rats with chronic renal failure (CRF), and to explore whether Cordyceps baoshen granule can improve hyperlipidemia hypercoagulability by regulating serum lipid and homocysteine levels in rats. Methods: after weighing 78 male SD rats, they were randomly divided into control group (n = 26), model group (n = 26) and cordyceps baoshen granule group (n = 26). The model group and Cordyceps group were treated with 5 / 6 nephrectomy to make CRF model. After the model was made, each group was given intragastric administration once a day for 12 weeks. The control group and the model group were given the same amount of distilled water as the Cordyceps baoshen granule decoction. The general state of rats was observed at 812 weeks after modeling, and the levels of serum creatinine (SCR), urea nitrogen (bun), cholesterol (Chol), triglyceride (TG), low density lipoprotein (LDL-C) and homocysteine (HCy) were measured in 24 hours urine protein quantity and serum levels of creatinine (SCR), urea nitrogen (bun), cholesterol (Chol), low density lipoprotein (LDL-C) and homocysteine (HCy). At the same time, 6 rats in each group were killed and the changes of renal histomorphology were observed by HE staining. The result is 1: 1. Cordyceps baoshen granule can effectively improve the general state of rats. Compared with the model group, Cordyceps baoshen granule could decrease the level of serum Scr-BUNand and proteinuria in CRF rats (P0.01P 0.05). The level of LDL-C and Hcy in serum of CRF rats was significantly lower than that of model group (P0.01P 0.05). After treatment with Cordyceps baoshen granules, the degree of glomerulosclerosis, interstitial fibrosis and infiltration of inflammatory cells in renal tubules were reduced. Conclusion 1. Cordyceps baoshen granule can improve the general state of CRF rats and delay the progression of CRF. Cordyceps baoshen granule can reduce the level of serum Scrn bun and proteinuria in CRF rats, and delay the progression of CRF. Cordyceps sinensis Baoshen granule can improve the hyperlipidemia and hypercoagulability by lowering the serum levels of TGG LDL-C and Hcy in the later stage of renal failure. Cordyceps baoshen granule can reduce the degree of glomerulosclerosis and interstitial fibrosis in 5-6 nephrectomy rats.
