针刺对哮喘大鼠肺组织中Eotaxin、RANTES表达的影响

【摘要】:目的:探讨针刺对支气管哮喘大鼠肺组织中Eotaxin、RANTES表达的调控。方法:将24只SPF级SD大鼠随机分为空白组、哮喘模型组、哮喘模型针刺组,每组8只。各组分别给予相应干预后,采用HE染色法观察大鼠肺组织中病理变化,免疫荧光染色法检测肺组织中Eotaxin、RANTES的表达,ELISA检测大鼠BALF及血清中Eotaxin、RANTES表达的程度。结果:哮喘模型针刺组大鼠肺组织病理改变较哮喘模型组明显减轻,与空白组差别不明显;哮喘模型组Eotaxin、RANTES蛋白表达高于哮喘模型针刺组(P0.05);哮喘针刺组BALF及血清中Eotaxin、RANTES含量均低于哮喘模型组(P0.05),但与空白组差异不明显(P0.05)。结论:哮喘大鼠BALF及血清中Eotaxin、RANTES的表达水平明显增高;针刺治疗可以明显降低其表达水平,提示针刺可以通过调节Eotaxin、RANTES的表达来减轻气道炎症反应。
[Abstract]:Objective: to investigate the effect of acupuncture on the expression of Eotaxin,RANTES in lung tissues of asthmatic rats. Methods: 24 SPF SD rats were randomly divided into blank group and acupuncture group with 8 rats in each group. The pathological changes of lung tissue were observed by HE staining, the expression of Eotaxin,RANTES in lung tissue was detected by immunofluorescence staining and the expression of BALF and Eotaxin,RANTES in serum were detected by Elisa. Results: the pathological changes of lung tissue in the acupuncture group of asthma model were significantly less than those in the asthmatic model group, but there was no significant difference between the two groups. The expression of Eotaxin,RANTES protein in asthma model group was higher than that in asthma model acupuncture group (P0.05), and the content of BALF and serum Eotaxin,RANTES in asthma acupuncture group was lower than that in asthma model group (P0.05), but there was no significant difference between asthma model group and blank group (P0.05). Conclusion: the expression of Eotaxin,RANTES in BALF and serum of asthmatic rats was significantly increased, and acupuncture treatment could significantly reduce the expression of Eotaxin,RANTES, suggesting that acupuncture can alleviate airway inflammation by regulating the expression of Eotaxin,RANTES.
