藏药陆额对黄嘌呤氧化酶活性的影响及其化学成分研究

【摘要】:陆额(Corydalis pallida)为罂粟科植物齿苞黄堇的全草,藏党参(Codonopsis nervosa)为党参属植物脉花党参的全草或者地上部分。陆额和藏党参是藏药罗堆多吉颗粒的主要组成部分,藏药罗堆多吉颗粒主要用于痛风病的治疗。关于陆额和藏党参对于黄嘌呤氧化酶活性的影响及陆额的化学成分尚未见报道。因此,本文选择陆额和藏党参为研究对象,研究其对黄嘌呤氧化酶活性的影响以及陆额化学成分的鉴定,为进一步探索藏药治疗痛风病的物质基础及作用机制打下基础。具体包括以下几个方面:1、藏党参对黄嘌呤氧化酶活性的影响目的:研究藏党参提取物及其单体对黄嘌呤氧化酶活性的影响。方法:制备藏党参的甲醇提取液,旋蒸并且干燥。将藏党参的提取物以及单体以缓冲液稀释为不同的浓度,采用紫外分光光度法测定对黄嘌呤氧化酶活性的影响。结果:藏党参的甲醇提取物及其分离纯化的单体对黄嘌呤氧化酶均有抑制作用并且呈现了良好的量效关系。2、罗堆多吉颗粒及其组方成分陆额对黄嘌呤氧化酶活性的影响目的:探讨罗堆多吉颗粒、陆额在不同浓度下对黄嘌呤氧化酶活性影响。方法:分别制备罗堆多吉颗粒的甲醇提取物和陆额的甲醇、丙酮以及水提取物,用缓冲液稀释成不同的浓度,再以荔枝草为阳性对照,采用紫外分光光度法测定各个提取物对黄嘌呤氧化酶活性的影响。结果:罗堆多吉颗粒甲醇提取物及陆额的各溶剂提取物对黄嘌呤氧化酶均具有明显抑制作用。3、陆额化学成分的分离与鉴定目的:研究陆额的化学成分。方法:称取干燥药材20Kg,经回流提取得到干浸膏。将干浸膏使用不同的溶剂进行萃取,得到极性不同的部位的浸膏。不同极性部位的浸膏,利用薄层分析,硅胶柱等色谱手段进行分离,得到不同的化合物。根据波谱分析鉴定化合物结构。结果:从陆额的乙酸乙酯和二氯甲烷部位分离提取13个成分。结论:13种化合物均为首次从该种植物得到。
[Abstract]:Land (Corydalis pallida) is the whole grass of Corydalis davidifolia and (Codonopsis nervosa) is the whole grass or the aboveground part of Codonopsis pilosula. Luzhang and Tibetan Codonopsis pilosula are the main components of Tibetan medicine Luoduoji granule, which is mainly used in the treatment of gout. No report has been reported on the effect of land and Tibetan Codonopsis pilosula on xanthine oxidase activity. Therefore, in this paper, the effects on xanthine oxidase activity and the identification of the chemical constituents of Xanthine oxidase were studied, which laid a foundation for further exploring the material basis and mechanism of Tibetan medicine in treating gout. The effects of Tibetan Codonopsis pilosula on xanthine oxidase activity objective: to study the effects of extracts and monomers of Tibetan Codonopsis pilosula on xanthine oxidase activity. Methods: the methanol extract of Codonopsis pilosula was prepared, steamed and dried. The extracts and monomers of Codonopsis pilosula were diluted to different concentrations in buffer solution, and the effect on xanthine oxidase activity was determined by ultraviolet spectrophotometry. Results: the methanol extract of Codonopsis pilosula and the monomers isolated and purified from Codonopsis pilosula had inhibitory effect on xanthine oxidase and showed a good dose-effect relationship. Effect of Luoduoji granule and its ingredients on xanthine oxidase activity objective: to study the effect of Luoduoji granule on xanthine oxidase activity at different concentrations. Methods: the methanol extract and the surface methanol, acetone and water extracts of Luopidoji granules were prepared, and then diluted into different concentrations with buffer solution, and the lychee grass was used as positive control. The effect of various extracts on xanthine oxidase activity was determined by UV spectrophotometry. Results: the methanol extract of Luopidoogi granules and the solvent extracts of the land surface had obvious inhibitory effects on xanthine oxidase. 3. Objective: to study the chemical constituents of the land surface. Methods: 20 kg dry medicinal materials were extracted by reflux extraction to get dry extract. The dry extract was extracted with different solvents to obtain the extract with different polarity. Different compounds were obtained by TLC and silica gel column chromatography. The structure of the compound was identified by spectral analysis. Results: 13 constituents were isolated from ethyl acetate and dichloromethane. Conclusion: all 13 compounds were obtained from this plant for the first time.
