针刺对去势雌兔干眼症模型眼表粘蛋白MUC5AC、MUC19表达的影响

【摘要】:目的研究针刺对更年期干眼症模型去势雌兔眼表蛋白MUC5AC和MUC19的表达影响,为更年期干眼症治疗提供依据。方法选择健康成年雌兔45只,将其随机分为对照组、模型组、针刺组(每组15只),模型组和针刺组行双侧卵巢切除建立干眼模型,2周后对针刺组采取针刺治疗,分别于去势前、2周后、3周后、6周后行Schirmer试验、泪膜破裂时间检测、荧光染色评分、虎红染色评分、免疫组化染色检测MUC5AC的MUC19表达水平。结果 2周和3周时模型组和针刺组Shirmer值、BUT时间、荧光染色评分、虎红染色评分值与对照组相比,差异有统计学意义(P0.05);3周时针刺组和模型组的Shirmer值和虎红染色评分差异有统计学意义(P0.05);6周时模型组的Shirmer值、BUT时间、荧光染色评分、虎红染色评分值与对照组和针刺组相比,差异有统计学意义(P0.05)。实验3周后针刺组MUC5AC和MUC19表达水平较模型组显著升高(P0.05),与对照组相比差异有统计学意义(P0.05);6周后针刺组黏蛋白表达水平继续升高,明显高于模型组(P0.05)。结论针刺治疗可提高眼表黏蛋白的表达,提示其在干眼症治疗中具有应用价值。
[Abstract]:Objective to study the effect of acupuncture on the expression of surface protein MUC5AC and MUC19 in ovariectomized female rabbits, and to provide evidence for the treatment of menopausal xerophthalmia. Methods Forty-five healthy adult female rabbits were randomly divided into control group, model group, acupuncture group (15 rabbits in each group), model group and acupuncture group with bilateral ovariectomies to establish dry eye model, and acupuncture group was treated with acupuncture 2 weeks later. Schirmer test was performed before castration, 2 weeks later, 3 weeks later and 6 weeks later. Tear film rupture time, fluorescent staining score, tiger red staining score and MUC19 expression level of MUC5AC were detected by immunohistochemical staining. Results the Shirmer, BUT time, fluorescent staining score and tiger red staining score of model group and acupuncture group were significantly different from those of control group at 2 and 3 weeks (P0.05). There were significant differences in Shirmer value and tiger red staining score between the acupuncture group and the model group at 3 weeks (P0.05). At 6 weeks, the Shirmer value, BUT time, fluorescent staining score and tiger red staining score of the model group were significantly different from those of the control group and the acupuncture group (P0.05). After 3 weeks, the expression of MUC5AC and MUC19 in the acupuncture group was significantly higher than that in the model group (P0.05), and the difference was statistically significant compared with the control group (P0.05). After 6 weeks, the expression of mucin in acupuncture group continued to increase, which was significantly higher than that in model group (P0.05). Conclusion Acupuncture therapy can improve the expression of mucin on ocular surface, suggesting that acupuncture can be used in the treatment of xerophthalmia.
