藏药莪达夏改善大鼠心肌缺血再灌注损伤入血活性成分分析研究

【摘要】:目的:本文通过实验对藏药莪达夏不同部位体外和体内化合物信息及其在大鼠体内可能的移行变化进行探讨研究。方法:通过醇提,萃取等方法提取得到藏药莪达夏乙酸乙酯部位、氯仿部位、水部位和正丁醇部位,应用LC-MS技术检测分析其化学成分,确定其化合物信息;将已适应饲养7天的大鼠随机分为4组:分别为乙酸乙酯部位组(EAEO)、氯仿部位组(CHEO)、正丁醇部位组(ZDCO)、水部位组(EDX-S),每组6只,各组大鼠禁食12小时后,分别于末次灌胃0、30、60、90、120、180、240、360、480、720、1 440 min眼眶静脉取血0.5 mL,应用LC-MS技术检测分析其化学成分,确定化合物信息,研究其在大鼠血浆中的移行变化;末次采血后采集大鼠肝脏和肠内粪便样品,进行肝微粒体转化实验和肠内转化实验,分别应用LC-MS技术检测分析其化学成分,确定化合物信息,研究其在大鼠肝脏和肠内的移行变化。结果:(1)通过LC-MS检测,得到莪达夏不同部位入血化学成分,其中:莪达夏氯仿部位有15个化学成分(包括12个黄酮类化合物,1个糖苷类化合物,2个其它类化合物);乙酸乙酯部位有18个化学成分(包括13个黄酮类化合物,1个酰胺类化合物,1个糖苷类化合物,3个其它类化合物);水部位有40个化学成分(包括24个黄酮类成分,2个酰胺类化合物,1个糖苷类化合物,13个其它类化合物);正丁醇部位有14个化学成分(包括10个黄酮类化合物,1个酰胺类化合物,3个其它类化合物)。(2)莪达夏不同部位中的一些黄酮类化合物(包括7-羟基黄酮,白杨素,黄芹素,槲皮素,7-甲氧基黄酮,鼠李素,2’-羟基-4’-甲氧基查尔酮,7-羟基二氢黄酮,2’,4’-二羟基查耳酮,染料木黄酮)可能在大鼠血浆中参与一系列生化反应,参与的多为Ⅰ相反应(包括羟基化反应和甲基化反应等)。(3)莪达夏不同部位中的一些黄酮类化合物(包括杨梅素,2’,4’-二羟基查尔酮,3’,4’,5,7-四羟基双氢黄酮)可能参与大鼠一系列肝脏代谢反应,参与的多为Ⅰ相反应(包括羟基化反应和甲基化反应等)。(4)莪达夏不同部位中的一些黄酮类化合物(包括槲皮素,7-甲氧基黄酮,7-羟基二氢黄酮,杨梅素)可能在大鼠体内参与一系列肠道代谢过程,参与的多为Ⅰ相反应(包括羟基化反应和甲基化反应等)。结论:1.莪达夏不同部位中参与体内代谢过程的化合物主要是黄酮类化合物。因此,认为总黄酮可能是莪达夏在大鼠体内发挥其药理和药效作用的主要有效成分。2.藏药莪达夏中黄酮类化合物参与体内代谢的途径可能主要是Ⅰ相反应,包括甲基化和羟基化反应等。3.参与体内代谢的黄酮类化合物包括杨梅素,白杨素,黄芹素,槲皮素,鼠李素,染料木黄酮,7-羟基黄酮,7-甲氧基黄酮,7-羟基二氢黄酮,2’,4’-二羟基查耳酮,2’-羟基-4’-甲氧基查尔酮和3’,4’,5,7-四羟基双氢黄酮。
[Abstract]:Aim: to study the information of compounds in different parts of Tibetan medicine Erdaxia in vitro and in vivo and their possible transference changes in rats. Methods: the components of ethyl acetate, chloroform, water and n-butanol of Tibetan medicine were extracted by alcohol extraction and extraction. The chemical constituents were detected and analyzed by LC-MS technique, and the information of their compounds was determined. The rats who had been fed for 7 days were randomly divided into 4 groups: ethyl acetate group, (EAEO), chloroform fraction group, (CHEO), n-butanol fraction group, (ZDCO), water fraction group (EDX-S), 6 rats in each group. The rats in each group were fasted for 12 hours. The chemical constituents were detected and analyzed by LC-MS for 0.5 mL, after the last intragastric infusion of the blood collected from the orbital vein at the last time. The chemical constituents were determined and the changes of the compounds in the plasma of the rats were studied by using the LC-MS technique. The results were as follows: (1) the changes of the chemical composition in the plasma of the rats were determined by using the LC-MS technique, and the chemical constituents were determined by the LC-MS technique. The changes of the chemical constituents in the plasma were studied. After the last blood collection, liver and intestinal fecal samples were collected, liver microsomal transformation test and enteral transformation test were carried out. The chemical constituents were detected and analyzed by LC-MS technique, and the compound information was determined. To study the changes of liver and intestine migration in rats. Results: (1) the chemical constituents of different parts of Edaraxia were obtained by LC-MS detection. Among them, there were 15 chemical constituents (including 12 flavonoids, 1 glycoside) in the chloroform of Edaraxia. 2 other compounds); There are 18 chemical constituents in ethyl acetate (including 13 flavonoids, 1 amide, 1 glycoside and 3 other compounds). There were 40 chemical constituents (including 24 flavonoids, 2 amides, 1 glycoside and 13 other compounds). There are 14 chemical constituents of n-butanol (including 10 flavonoids, 1 amides, 3 other compounds). (2). Yellow apigenin, quercetin, 7-methoxyflavone, rohamnetin, 2-hydroxy-4- methoxy chalcone, 7- hydroxy dihydroflavone, 2-dihydroxy chalone, Genistein may be involved in a series of biochemical reactions in rat plasma, mostly in phase I reactions (including hydroxylation reaction and methylation reaction), and some flavonoids (including myricetin) in different parts of Erdaxia. (2) it may be involved in a series of metabolic reactions in the liver of rats.
