悬灸治疗中性粒细胞哮喘的细胞免疫学机制研究

【摘要】：目的:探讨悬灸对中性粒细胞哮喘相关信号因子的影响。方法:SD大鼠随机分为正常组、模型组、地塞米松组与悬灸组,每组8只。用鼻腔滴入脂多糖、腹腔注射卵白蛋白及雾化吸入卵白蛋白引喘制备中性粒细胞哮喘模型。造模成功后,地塞米松组按0.5mg/kg予以腹腔注射地塞米松,悬灸组悬灸"肺俞""脾俞""肾俞""膻中",40min/次,均每日1次,共14d。治疗结束后,统计各组支气管肺泡灌洗液(BALF)白细胞计数与分类,流式细胞术检测脾及外周血辅助性T淋巴细胞17(Th 17)含量,酶联免疫法检测BALF中的白介素17(IL-17)含量,免疫组化法及免疫印迹法检测右肺下叶维甲酸相关孤儿受体γ的胸腺异构体(ROR-γt)蛋白的表达水平,real-time PCR法检测ROR-γt mRNA的表达水平。结果:模型组与正常组比较,气道有大量炎性细胞浸润,中性粒细胞显著升高(P0.01),BALF中IL-17含量显著增高(P0.01),脾与外周血中Th 17含量显著增多(P0.01),肺组织中ROR-γt蛋白与mRNA的表达水平显著升高(P0.01)。地塞米松组与模型组比较,上述指标皆显著降低(P0.01);悬灸组与模型组比较,上述指标亦显著降低(P0.05,P0.01)。与悬灸组比较,地塞米松组外周血Th 17含量更低(P0.05)。结论:悬灸可以通过抑制ROR-γt转录因子表达,下调炎性反应因子IL-17含量,降低气道内中性粒细胞水平,从而达到减轻中性粒细胞哮喘炎性反应的作用,其疗效与地塞米松相当。
[Abstract]:Objective: to investigate the effect of moxibustion on signal factors related to neutrophil asthma. Methods: SD rats were randomly divided into normal group, model group, dexamethasone group and moxibustion group with 8 rats in each group. Neutrophil asthma model was induced by nasal drip of lipopolysaccharide, intraperitoneal injection of ovalbumin and aerosol inhalation of ovalbumin to induce asthma. After the model was successfully established, dexamethasone was injected intraperitoneally according to 0.5mg/kg, and suspended moxibustion group was moxibustion of "Feshu", "spleen Shu", "Shenshu" and "Shanzhong", 40min/ times, once a day for 14 days. After treatment, the count and classification of (BALF) leukocytes in bronchoalveolar lavage fluid (BALF) were counted, and the contents of helper T lymphocyte 17 (Th 17) in spleen and peripheral blood were detected by flow cytometry. The content of interleukin 17 (IL-17) in BALF was detected by enzyme linked immunosorbent assay (Elisa), and the expression of ROR- 纬 t protein in the thymus isomer (ROR- 纬 t) of retinoic acid-associated orphan receptor 纬 in the lower lobe of the right lung was detected by immunohistochemistry and Western blotting. The expression of ROR- 纬 t mRNA was detected by real-time PCR method. Results: compared with the normal group, there were a lot of inflammatory cells infiltrating the airway, and the neutrophils were significantly increased (P0.01), BALF, P0.01), spleen and peripheral blood (P0.01), the content of Th 17 in spleen and peripheral blood was significantly increased (P0.01). The expression of ROR- 纬 t protein and mRNA was significantly increased in lung tissue (P0.01). Compared with the model group, the above indexes in the dexamethasone group were significantly lower than those in the model group (P0.01), and those in the moxibustion group were significantly lower than those in the model group (P0.05, P0.01). Compared with the moxibustion group, the content of Th 17 in peripheral blood of dexamethasone group was lower than that of moxibustion group (P0.05). Conclusion: moxibustion can inhibit the expression of ROR- 纬 t transcription factor, down-regulate the content of inflammatory response factor (IL-17) and decrease the level of neutrophil in airway, so as to alleviate the inflammatory response of neutrophil asthma. Its curative effect is equivalent to that of dexamethasone.
