芪桂颗粒对甲状腺功能减退大鼠IL-1及IFN-γ的影响

【摘要】:目的:本实验通过观察自拟方芪桂颗粒对甲减大鼠血清IL-1及IFN-γ表达水平影响,以观察芪桂颗粒治疗甲减的治疗效果,探讨其对下丘脑-垂体-甲状腺轴及相关细胞因子的调控作用,并阐明其作用机理。方法:选取健康清洁级Wistar大鼠50只,随机分为空白组、模型组、西药组、中药低剂量组和中药高剂量组5组,每组10只。空白组给予生理盐水灌胃,其余各组均给予0.1%丙硫氧嘧啶(Propylthiouracil,PTU)生理盐水溶液灌胃,造甲减大鼠模型。15天后采血检测甲状腺功能及IL-1、IFN-γ含量,证实模型建立成功,后给予相应的西药、芪桂颗粒连续治疗8周。8周后统一采血,采用电化学发光法检测FT3、FT4、TSH、TG-Ab、TPO-Ab。采用ELISA试剂盒检测IL-1、IFN-γ。结果:造模后的甲减大鼠在给药治疗后,其甲减症状均有不同程度的改善。给药8周之后,中药各剂量组FT3、FT4值均高于模型组,有显著性差异(P0.05);中药各剂量组TSH值均低于模型组,有显著性差异(P0.05);中药高剂量组的FT3、FT4、TSH值与中药低剂量组比较,有极显著性差异(P0.01);中药高剂量组的FT3、FT4、TSH值与西药组比较,无统计学差异(P0.05)。中药低、高剂量组IL-1、IFN-γ均明显低于模型组,有显著性差异(P0.05);中药低剂量组与西药组IL-1、IFN-γ值比较,无统计学差异(P0.05);中药高剂量组与空白组IL-1、IFN-γ值比较,无统计学差异(P0.05);中药高剂量组与西药组IL-1、IFN-γ值比较,有极显著性差异(P0.01)。给药后,各组大鼠血清TG-Ab、TPO-Ab改善不显著,无统计学差异(P0.05)。结论:芪桂颗粒可以显著改善甲减大鼠一般状态,升高FT3、FT4水平,降低TSH水平。芪桂颗粒高剂量组可以显著降低IL-1、IFN-γ水平,改善大鼠甲减症状。
[Abstract]:Objective: to observe the effect of Qigi granule on the expression of IL-1 and IFN- 纬 in serum of hypothyroidism rats. To investigate the regulation of hypothalamus-pituitary-thyroid axis and related cytokines, and to elucidate its mechanism. Methods: 50 healthy and clean grade Wistar rats were randomly divided into blank group, model group, western medicine group, low dose Chinese medicine group and high dose Chinese medicine group with 10 rats in each group. The rats in the blank group were given normal saline and the other groups were given 0.1% Propylthiouracil,PTU saline solution to make the model of hypothyroidism. After 15 days, the thyroid function and the content of IL-1,IFN- 纬 were measured. It was proved that the model was established successfully, and then the corresponding western medicine was given. Qigui granule was treated continuously for 8 weeks. After 8 weeks, blood was collected uniformly, and FT3,FT4,TSH,TG-Ab,TPO-Ab. was detected by electrochemiluminescence method. ELISA kit was used to detect IL-1,IFN- 纬. Results: the hypothyroidism symptoms of hypothyroidism rats were improved in varying degrees. After 8 weeks of administration, the FT3,FT4 value of each dose group was higher than that of the model group (P0.05), and the TSH value of each dose group was lower than that of the model group (P0.05). The FT3,FT4,TSH value of the high dose group was significantly higher than that of the low dose group (P0.01), while the FT3,FT4,TSH value of the high dose group was not significantly different from that of the western medicine group (P0.05). IL-1,IFN- 纬 in low and high dose group was significantly lower than that in model group (P0.05), but there was no significant difference in IL-1,IFN- 纬 between low dose group and western medicine group (P0.05). There was no significant difference in IL-1,IFN- 纬 between the high dose group and the blank group (P0.05); the IL-1,IFN- 纬 value in the high dose group was significantly higher than that in the western medicine group (P0.01). After administration of the drug, the improvement of serum TG-Ab,TPO-Ab in each group was not significant, there was no statistical difference (P0.05). Conclusion: Qigui granule can significantly improve the general state of hypothyroidism rats, increase the level of FT3,FT4 and decrease the level of TSH. Qigui granule high dose group can significantly reduce the level of IL-1,IFN- 纬, improve the symptoms of hypothyroidism in rats.
