基于DNA条形码技术的维吾尔药材桑葚基原鉴定研究

【摘要】:该研究通过分析多途径来源的新疆桑属植物及药材样本的ITS2,psbA-trnH序列,为药材分子鉴定提供依据。以51个新疆桑属植物及药材为样本,对其ITS2,psbA-trnH序列进行PCR扩增和测序,用MEGA 6.0计算其种内、种间Kimura 2-parameter(K2P)距离,分析变异位点,并构建NJ鉴别树。ITS2序列分析结果显示,桑Morus alba、鞑靼桑M.alba var.tatarica、黑桑M.nigra种内无变异;桑与鞑靼桑种间无变异;桑与黑桑种间存在13个变异位点,种间平均KP2遗传距离为0.04;桑与药材样本间无信息变异位点,NJ鉴别树可将桑及鞑靼桑与黑桑区分。psbA-trnH序列分析结果显示,桑与黑桑种内各有1个变异位点,3种植物种间存在插入/缺失变异,可相互区分;种间变异与药材样本内变异一致。因此,ITS2序列可将来源于桑、鞑靼桑的药材样本与黑桑区分,psbA-trnH序列可将来源于三者的药材样本区分,为维吾尔药材真伪鉴别及市场监管提供依据。
[Abstract]:In this study, the ITS2,psbA-trnH sequences of mulberry plants and their samples from various sources were analyzed to provide basis for molecular identification of medicinal materials. The ITS2,psbA-trnH sequences of 51 mulberry plants and medicinal materials from Xinjiang were amplified by PCR and sequenced. The distance of Kimura 2-parameter (K2P) between species and species was calculated by MEGA 6.0, and the variation sites were analyzed. NJ identification tree was constructed. The results of ITS2 sequence analysis showed that there was no variation in the M.nigra species of Morus alba, Tatar mulberry M.alba var.tatarica,. There were 13 variation sites between mulberry and black mulberry, and the average genetic distance of KP2 between mulberry and black mulberry was 0.04. There were no information mutation sites between mulberry and medicinal material samples, and NJ discriminant tree could distinguish mulberry and Tartar mulberry from black mulberry. The results of psbA-trnH sequence analysis showed that there was one mutation locus in mulberry and black mulberry species, and there was insertion / deletion mutation among planting species. Distinguishable from each other; The interspecific variation was consistent with the variation in the samples of medicinal materials. Therefore, ITS2 sequence can distinguish the samples from mulberry, Tartar mulberry and black mulberry, psbA-trnH sequence can distinguish the samples from the three, and provide the basis for the identification and market supervision of Uygur medicinal materials.
