电针俞募穴对肠动力亢进大鼠海马组织BDNF表达的影响

【摘要】:目的:探讨电针大肠俞募穴治疗肠动力异常亢进的作用机制。方法:60只大鼠随机分为对照组、模型组、俞募组、俞穴组、募穴组,每组12只。采用番泻叶煎剂灌胃造模法,同日电针刺激天枢、大肠俞,每次20min,共治疗10次。运用光镜、HE染色、免疫组化等技术,观察电针治疗后大鼠排便情况、海马及肠道组织细胞形态学改变、海马组织中BDNF蛋白表达水平。结果:肠动力亢进模型建立后,与对照组比较,1 h排便粪粒数、24 h排便粪粒总数、24 h粪便含水量均显著增高(P0.01),海马CA2区、CA3区、DG区BDNF蛋白表达水平显著减少,以DG区最为明显(P0.01);治疗后,募穴组大鼠的1 h排便粪粒数、募穴组和俞募组大鼠的24 h排便粪粒总数和24 h粪便含水量较模型组显著降低(P0.01),俞募组、募穴组、俞穴组上述脑区中BDNF蛋白表达水平显著回升,以DG区最为明显(P0.01);光镜下模型组、针刺组大鼠海马、肠道组织形态均有所改变,针刺组改变程度较轻。结论:大肠俞募穴可以改善肠动力亢进大鼠的异常排便情况,通过提高海马DG区BDNF蛋白表达水平而发挥中枢调控作用可能是其起效的一个重要机制。
[Abstract]:Objective: to explore the mechanism of electro-acupuncture (EA) at large intestine Shu (EA) for treatment of abnormal hyperactivity of intestinal motility. Methods: 60 rats were randomly divided into three groups: control group, model group, Yu exixie group, Yu acupoint group and Baixue point group, with 12 rats in each group. The model was made by using Senna leaf decoction by gavage. Electroacupuncture was used to stimulate Tianshu and large intestine on the same day for 20 minutes each time for 10 times. By means of light microscopy, HE staining and immunohistochemistry, the defecation, morphological changes of hippocampal and intestinal tissue cells and the expression of BDNF protein in hippocampus were observed after electroacupuncture treatment in rats. Results: compared with the control group, the number of feces, the total number of feces at 24 h and the water content of feces at 24 h after the establishment of the model were significantly higher than those in the control group (P0.01). The number of fecal granules in hippocampal CA2 area and CA3 area were significantly higher than those in the control group (P0.01). The expression level of BDNF protein in DG region was significantly decreased, especially in DG region (P0.01). After treatment, the number of fecal particles at 1 hour in group A, the total number of fecal particles in 24 h and water content in 24 h in group A and group Yu were significantly lower than those in the model group (P0.01), and those in group Yu and Yu were significantly lower than those in group A (P 0.01), and the number of fecal particles in group A was significantly lower than that in group A. The expression level of BDNF protein in the above brain regions increased significantly in the Yu-acupoint group, especially in the DG area (P0.01). The histological changes of hippocampus and intestinal tract of rats in model group and acupuncture group were all changed under light microscope, and the degree of change in acupuncture group was less severe than that in control group. Conclusion: Jiaoshu Xizhi point can improve the abnormal defecation of rats with intestinal motility hyperkinesia, and the central regulatory role may be an important mechanism of its effect by increasing the expression of BDNF protein in the DG region of hippocampus.
