壮药篱栏网质量标准研究

【摘要】:目的:对壮药篱栏网进行真伪、优劣评价,并初步建立壮药篱栏网药材质量标准。方法:(1)通过基源鉴定、性状鉴定、显微鉴定与理化鉴定系统地对篱栏网进行生药学定性研究。(2)照《中华人民共和国药典》2015年版四部中规定的方法对篱栏网药材进行限量检查,包括水分、总灰分、酸不溶性灰分、浸出物等。(3)按照中药化学预实验方法对3个产地篱栏网进行成分预试。(4)以芦丁为对照品,采用紫外-可见光分光光度法对广西12个批次篱栏网药材进行总黄酮含量测定,确定其所含总黄酮的限量。(5)以绿原酸为对照品,采用高效液相色谱法对广西12个批次篱栏网药材进行绿原酸含量测定,以确定其所含绿原酸的限量。(6)基于抗菌效价的生物检定法检测篱栏网药材的生物活性。结果:(1)基源、性状、显微及薄层色谱鉴别均具显著性特征,可作为鉴别篱栏网药材真伪的依据。(2)实验结果表明,篱栏网药材水分含量范围为6.26%~9.96%,初步拟定其水分不得超过11.00%;总灰分含量范围为6.31~11.55%,初步拟定总灰分不得超过13.00%;酸不溶性灰分含量范围为0.20%~1.60%,初步拟定酸不溶性灰分不得超过1.70%;浸出物含量在4.69%~9.22%之间,初步拟定醇溶性浸出物含量不得少于4.00%。(3)化学成分预实验显示篱栏网中含有黄酮类、糖、多糖、苷类、生物碱、蒽醌类、蛋白质、氨基酸、有机酸及鞣质或酚类等化学成分类型。(4)广西12批次的篱栏网药材总黄酮含量测得值在5.13%~14.32%之间,初步拟定其所含总黄酮不得少于4.00%。(5)广西12批次篱栏网药材中绿原酸含量测得值在0.22%~0.64%之间,初步拟定其所含绿原酸(C16H18O9)不得少于0.20%。(6)篱栏网药材的对数剂量和抑菌效应呈线性关系,相关性好,基于抗菌效价的生物检定法可行,初步拟定篱栏网(0.5g·ml-1)的效价不得低于0.8000U/ml。结论:为篱栏网药材的准确鉴定和质量标准的制定提供了科学依据,也为篱栏网的开发应用研究奠定了基础。
