傣药肾茶的迷迭香酸和咖啡酸含量测定及指纹图谱研究

【摘要】:目的:建立不同产地15批肾茶药材的含量测定方法及指纹图谱,为肾茶的质量控制提供依据。方法:采用RP-HPLC法,色谱柱:Unltary-C_(18)100A(250mm×4.6mm,5μm),流动相:甲醇-0.1%甲酸溶液,梯度洗脱,洗脱时间:85min,流速:1mL/min,检测波长:254nm,柱温:30℃。不同产地肾茶相似度评价采用2004A版中药色谱指纹图谱相似度评价系统软件,以SPSS 20.0软件对肾茶不同产地进行聚类分析。结果:对15批不同产地的肾茶进行了含量测定和相似度分析,其相似度在0.874-1.000之间,同时通过比对原儿茶醛、咖啡酸、迷迭香酸对照品的保留时间归属了5、8、20号峰分别为原儿茶醛、咖啡酸、迷迭香酸,在此基础上建立了肾茶的HPLC指纹图谱,肾茶不同产地的聚类分析显示云南产的肾茶质量要普遍优于其他产地,尤其是云南西双版纳产的,其迷迭香酸含量最高可达到1.31%。结论:建立的方法科学,结果可靠,简便可行,可以达到对肾茶质量的有效控制。
