毛细管电泳pH酸修饰在线富集方法检测唾液中鸦片类毒品

【摘要】:建立了唾液中鸦片类毒品吗啡、可待因和6-单乙酰吗啡的毛细管电泳pH酸修饰在线富集方法,并进行了方法学考察。100mmol/L的磷酸盐缓冲液(含20%甲醇和5%异丙醇)作为背景缓冲液,10kV下电迁移进样99s,然后电迁移(2kV,2s)引入一段100mmol/L的盐酸区带,最后施加25kV电压进行分离检测。3种毒品在20~1 000ng/mL范围内具有良好的线性关系,r为0.9934~0.9998,检出限均为7ng/mL,相对标准偏差(RSD)19%(n=5)。该方法样品前处理简单,只需用甲醇稀释样品便可进样分析,方法灵敏度较高、稳定性较好,可用于生物检材中痕量鸦片类毒品的检验。
[Abstract]:A method for on-line enrichment of opioid drugs morphine, codeine and 6-monoacetylmorphine in saliva by capillary electrophoresis with pH acid modification was established. The phosphate buffer of 100mmol/L (containing 20% methanol and 5% isopropanol) was used as the background buffer. The electromigration of 10kV was carried out for 99s, and then the electromigration (2kV / 2s) was introduced into the hydrochloric acid zone of 100mmol/L. Finally, the 25kV voltage was applied to separate the three drugs. The linear relationship of the three drugs was obtained in the range of 20 000ng/mL (r = 0.9934 卤0.9998), the detection limit was 7ng / mL, and the relative standard deviation (RSD) was 19% (n ~ (5). The method is simple in sample pretreatment and can be analyzed by dilution of methanol. The method has high sensitivity and good stability. It can be used for the detection of trace opiates in biological samples.
