福尔马林固定液和保存温度对肝脏中地西泮稳定性的影响

【摘要】: 目的建立福尔马林溶液固定的家兔肝脏中地西泮的检测方法。研究不同浓度不同酸度福尔马林固定液和保存温度对肝脏中地西泮稳定性的影响,为地西泮中毒的法医学鉴定提供相关依据。 方法1.福尔马林固定液对家兔肝脏中地西泮稳定性的影响:家兔9只随机分成3组(即5%、10%、20%福尔马林溶液组),每组3只,家兔分别以地西泮704.3 mg/kg剂量灌胃,2h后处死,每只家兔取肝脏组织85份,0.5g/份,即刻用乙醚提取5份肝脏中的地西泮,其余样品分别浸泡于4ml酸性、中性、碱性福尔马林溶液和水溶液中,不同时间点(1d、7d、14d、28d)提取,气相色谱法检测固定的肝脏及固定液中地西泮含量。2.保存温度对家兔肝脏中地西泮稳定性的影响:家兔分别以地西泮704.3 mg/kg剂量灌胃,2h后处死,每只家兔取肝脏组织42份,即刻用乙醚提取6份肝脏中的地西泮,其余样品分别保存于室温(18~24℃)、冷藏(4℃)和冷冻(-20℃)条件下,不同时间点(1d、7d、14d、28d)提取并检测其中地西泮含量。 结果1.地西泮在肝脏中的线性检测范围为0.1~64μg/ml,最低检出限为0.05μg/ml。2. 5%与20%福尔马林溶液固定肝脏中地西泮的相对含量呈下降趋势(P0.05),而10%福尔马林溶液固定的肝脏中地西泮的相对含量仅在酸性条件组呈下降趋势(P0.05),碱性条件组地西泮的相对含量无显著性变化(P0.05),中性条件组地西泮的相对含量呈先下降后上升的趋势(P0.05)。3. 10%福尔马林溶液组较5%与20%福尔马林溶液组肝脏中地西泮的相对含量更高。4.肝脏在室温、冷藏和冷冻条件下保存时,肝脏中地西泮含量均呈下降趋势,三个温度组间地西泮回收率变化存在显著性差异(P0.05)。 结论肝脏中地西泮在10%碱性福尔马林溶液中保存相对稳定,在5%和20%福尔马林溶液及10%中性和酸性福尔马林溶液中保存不稳定。储存温度和存放时间均影响样品中地西泮的含量,地西泮在冷冻条件下保存时稳定性相对较好。
[Abstract]:Objective to establish a method for the determination of diazepam in rabbit liver fixed with formalin solution. To study the effect of different concentration and acidity of formalin fixative solution and preservation temperature on the stability of diazepam in liver, and to provide relevant evidence for forensic identification of diazepam poisoning. Method 1. Effect of formalin fixative solution on the stability of diazepam in the liver of rabbits: nine rabbits were randomly divided into 3 groups, 3 in each group, with diazepam at a dose of 704.3 mg/kg. Two hours later, each rabbit was killed. 85 liver tissues and 0.5g/ were taken from each rabbit. Diazepam was extracted from 5 liver samples with ether immediately. The other samples were immersed in 4ml acidic, neutral and alkaline formalin solution and aqueous solution respectively. Diazepam was extracted at different time points (1 d ~ 7 d ~ 14 d ~ (28) d), and the content of diazepam in fixed liver and fixed solution was determined by gas chromatography. The effect of preservation temperature on the stability of diazepam in rabbit liver: diazepam was perfused by stomach with diazepam at a dose of 704.3 mg/kg. After 2 hours, 42 samples of liver tissue were taken from each rabbit and 6 samples of diazepam were extracted with ether. The other samples were stored at room temperature (1824 鈩,
