改良型乳腺癌淋巴结转移模型的制备及超声表现

【摘要】:目的现行方法建立的乳腺癌动物模型多用于观察其生物学表现,不适于探索新的淋巴结转移治疗方案。本研究通过改良制作方法建立新型的乳腺癌淋巴结转移模型,为探索新的治疗方法提供实验动物模型。材料与方法采用组织块包埋法将VX2传代瘤株种植于30只雌性兔左侧第3只乳头的乳垫内,每周采用触诊法和常规超声查看肿瘤及淋巴结生长情况。待触及肿大的淋巴结后切除乳腺癌组织。第4周随机摘取15枚淋巴结,采用病理学观察淋巴结形态及组织学改变。将游标卡尺与超声测量结果进行对照,比较其差异。结果 2周后,26只实验兔可触及乳腺内质韧的类圆形结节;超声呈低回声,边界欠清,内部及周边血流丰富。约3周后,23只实验兔的左侧腋窝处可触及肿大的淋巴结,病理证实为转移性淋巴结,淋巴结转移率为88.5%,造模成功率为76.7%。超声与病理两种方法测量淋巴结最大直径差异无统计学意义(P0.05)。结论改良法建立乳腺癌淋巴结转移模型周期短、成功率高、方法简单,能够为研究新疗法提供动物模型;常规超声能够实时监测肿瘤的生长状况,操作方便、快捷。
[Abstract]:Objective the animal models of breast cancer established by the present method are mostly used to observe their biological manifestations and are not suitable to explore a new treatment scheme for lymph node metastasis. In this study, a new model of lymph node metastasis of breast cancer was established by modified method. Materials and methods VX2 subculture was implanted into the breast pad of the third third nipple on the left side of 30 female rabbits by tissue mass embedding. The tumor and lymph node growth were examined by palpation and conventional ultrasound every week. 15 lymph nodes were randomly removed at 4 weeks after resection of breast cancer tissues after touching enlarged lymph nodes. The morphological and histological changes of lymph nodes were observed by pathology. The results of Vernier caliper were compared with the results of ultrasound. Results after 2 weeks, 26 rabbits could reach the ductile round nodules of mammary gland. The left axillary lymph nodes of 23 experimental rabbits were palpable at the left armpit about 3 weeks later. The lymph nodes were confirmed as metastatic lymph nodes by pathology. The lymph node metastasis rate was 88.5 and the success rate was 76.70.There was no significant difference between ultrasound and pathology in the measurement of lymph node maximum diameter. Conclusion the modified method has the advantages of short period, high success rate and simple method for establishing lymph node metastasis model of breast cancer. It can provide animal model for the study of new therapy, and conventional ultrasound can monitor tumor growth in real time, which is convenient and fast.
