全血标本离心前不同保存温度与时间对巨细胞病毒核酸检测结果的影响

【摘要】:目的探讨全血标本离心前不同温度与时间对人巨细胞病毒(human cytomegalovirus,HCMV)核酸检测(nucleic acid amplification test,NAT)结果的影响,以确定离心前放置的安全时限。方法采集含高、中、低DNA载量的全血。采血后按保存温度分为4℃、25℃、37℃组,各组放置0、1、6、12、24、48、72 h后进行离心,行实时荧光定量PCR。结果高浓度4℃组离心前放置72 h,25℃与37℃组离心前放置24 h,无明显变化;中浓度4℃组离心前放置24 h,25℃组、37℃组离心前放置12 h,无明显变化;低浓度4℃组、25℃组离心前放置12 h,37℃组离心前放置6 h,无明显变化。结论为保证HCMV NAT结果的准确性,全血标本最好在离心前4℃保存,并在采血后6 h内及时离心送检,高温条件下更应注意。
