广豆根黄酮类成分及黑升麻皂苷成分的研究

【摘要】:广豆根及黑升麻具有抗肿瘤、抗炎等药理功效,是重要的传统中药材。广豆根所含的黄酮类成分具有较好的抗溃疡活性和黑升麻所含的皂苷成分具有较好的缓解更年期作用。本文对广豆根的黄酮类成分和黑升麻皂苷成分进行研究,为药理活性和构效关系奠定基础。采用硅胶柱色谱、葡聚糖凝胶色谱Sephadex LH-20、反相柱色谱及高效液相色谱(High Performance Liquid Chromatography,HPLC)等色谱技术,并结合结晶与重结晶等分离手段,对广豆根进行分离纯化。采用质谱(Mass Spectrometry,MS)、核磁共振波谱(Nuclear Magnetic Resonance Spectroscopy,NMR)、紫外光谱(Ultraviolet Absorption Spectroscopy,UV)等现代波谱学技术,对分离到的化合物进行结构鉴定。采用色谱技术,从广豆根的二氯甲烷部分,分离到11个化合物。经波谱学技术,鉴定出9个黄酮类化合物:广豆根素(1,1.300 g)、多花胡枝子素A3(2,0.810 g)、L-高丽槐素(3,0.325 g)、光甘草酚(4,0.056 g)、广豆根酮(7,0.051 g)、山豆根色烯素(8,0.750 g)、Lupinifolin(9,0.015 g)、5-Hydroxysophoranone(10,0.18 g),同时还得到3个非黄酮类化合物:β-谷甾醇(5,0.098 g)羽扇豆醇(6,0.043 g)和大豆皂醇B(11,0.010 g)。化合物2为首次从该属植物中分离得到,化合物10为首次从该植物中分离得到。采用反复柱色谱和重结晶等方法对黑升麻的乙酸乙酯部位进行分离纯化,从黑升麻中分离得到黑升麻苷(0.208 g)和27位脱氧黑升麻苷(0.017 g),纯度分别为98.2%和98.3%,产率分别是0.020%和0.001%。
[Abstract]:Gangdou root and black cohosh have anti-tumor, anti-inflammatory and other pharmacological effects. It is an important traditional Chinese medicinal material. The flavonoids contained in the root of Guangdou have better anti-ulcer activity and the saponins contained in the black cohosh have a better effect on relieving climacteric. The components of saponins were studied. For the basis of pharmacological activity and structure-activity relationship, silica gel column chromatography and dextran gel chromatography (Sephadex LH-20) were used. Reversed phase column chromatography (RPC) and high performance liquid chromatography (HPLC) with high Performance Liquid chromatographyHPLC. In combination with crystallization and recrystallization, the root was separated and purified by mass spectrometric mass spectrometry (MS). Nuclear Magnetic Resonance spectroscopy (NMRs). Ultraviolet Absorption Spectroscopy (UV) and other modern spectroscopic techniques. 11 compounds were isolated from the dichloromethane part of Gangdou root by chromatographic technique, and 9 flavonoids were identified by spectroscopic technique. 1.300 g / g, 0.300 g / g, 0.810 g / L, 0.325 g / L, 0.056 g / L, 0.056 g / L, 7g / L, respectively. 0. 051 g / g, Lupinifolin9 (0. 015 g). 5-Hydroxysophoranoneum 100.18 g, and three non-flavonoids, 尾 -sitosterol 50.098 g) were also obtained. Compound 2 was isolated from this genus for the first time. Compound 10 was isolated from the plant for the first time. Repeated column chromatography and recrystallization were used to isolate and purify ethyl acetate from black cohosh. The purity was 98.2% and 98.3%, respectively. The yields were 0.020% and 0.001, respectively.
