野生沉香的鉴别方法

【摘要】:【目的】建立有效、准确、科学的野生沉香鉴别技术,为沉香的鉴定和品质评价体系建立提供参考依据。【方法】采用醇溶性浸出物含量、显色反应、薄层色谱分析(TLC)和高效液相色谱(HPLC)方法,对10批次野生沉香进行鉴别。【结果】10批次野生沉香的醇溶性浸出物含量整体较高,明显大于《中华人民共和国药典》(2015年版)规定的10.0%;野生沉香的显色反应除呈现人工沉香的樱红色或浅樱红外,还出现紫堇色、浅红色和浅紫色在内的多样性显色,可辅助鉴别野生沉香;薄层色谱操作简单,节省材料,且野生沉香样品在薄层鉴别中呈现清晰的荧光斑点和较好的分离效果,对于大部分野生沉香样品可通过薄层色谱进行基础鉴别;通过相似度评价、峰面积比较,建立10批次野生沉香的HPLC指纹图谱,并确定9个野生沉香共有特征峰,确定野生和人工沉香HPLC特征指纹图谱的差异性。【结论】基于各方法的优势及精准程度,本文提出在显微鉴别的基础上,通过醇溶性浸出物含量、显色反应、薄层色谱分析和HPLC的指纹图谱及特征峰鉴别野生沉香,可为沉香品质评价体系建立提供参考。
[Abstract]:[objective] to establish an effective, accurate and scientific identification technique for wild Amorpha sinensis, and to provide a reference for the identification and quality evaluation system. [methods] Alcohol-soluble extracts were used for color reaction. TLC (TLC) and HPLC (High performance liquid Chromatography) methods were used to identify 10 batches of wild Acanthia sinensis. [results] the alcohol-soluble extracts of 10 batches of wild Acanthia sinensis were higher in whole. It is obviously larger than the 10. 0 specified in the Pharmacopoeia of the people's Republic of China (2015 edition). The color reaction of wild Amomum sinensis is not only the fuchsia red or light cherry infrared, but also the color of Corydalis, light red and light purple. TLC was easy to operate and save materials, and the samples of wild Amorpha showed clear fluorescent spots and good separation effect in TLC identification. For most of the samples, TLC can be used for basic identification, and through similarity evaluation, peak area comparison, the HPLC fingerprint of 10 batches of wild Acanthia sinensis was established, and 9 common characteristic peaks were determined. [conclusion] based on the advantages and accuracy of each method, this paper proposed that the content of alcohol soluble extracts and color reaction should be used on the basis of microscopic identification. TLC, fingerprint and characteristic peak of HPLC can be used to identify wild Amorpha sinensis, which can provide reference for the establishment of quality evaluation system.
