基于罗丹明B结构修饰的pH荧光探针的合成及应用研究

【摘要】:目的:pH值在各种系统中起着举足轻重的作用,因此对pH值的检测是至关重要的。目前最常见的方法是采用玻璃电极来测定pH值,但是用玻璃电极测pH值时常常受到金属离子(如:钠、钾、锂)的干扰,导致测定结果不准确。与电化学方法相比,荧光探针的光学测量并不存在这些缺点。此外,荧光探针在选择性、灵敏性、纳米级空间分辨有着非常明显的优势。方法:我们以罗丹明B作为探针的基本骨架,连接不同结构的五原或六原杂环化合物合成目标产物,并用荧光分光光度计对其荧光性能进行研究,同时也研究了其细胞毒性和细胞成像性。结果:成功合成了荧光分子探针A、B、C、D、E,并通过1HNMR、13CNMR对其结构进行确认。探针D在Britton-Robinson(BR)缓冲溶液中随着p H由4.26增加到7.26其荧光强度逐渐减弱,溶液颜色也逐渐变淡直至消失。在可逆性实验中,当p H值在4.26和6.98来回变动时,荧光强度也可以恢复到原来的值,循环至少六次。在进行抗金属离子干扰能力实验时,不管pH值是在7.26还是4.26时金属离子对探针D的识别都几乎没有影响。而探针D在和PC12、MDA-MB-231细胞共培养时,低浓度的探针D对两种细胞的损伤都比较小,但令人遗憾的是探针D并没有能对细胞的溶酶体进行染色。结论:虽然在细胞成像上探针D没有达到令人满意的效果,但是其对质子的高选择性和高灵敏性可以应用于弱酸性环境监测。
[Abstract]:Objective: the pH value plays an important role in various systems, so the detection of pH value is very important.At present, the most common method is to use glass electrode to measure pH value. However, the measurement of pH value by glass electrode is often interfered by metal ions (such as sodium, potassium, lithium), which leads to inaccurate results.Compared with electrochemical method, the optical measurement of fluorescent probe does not have these disadvantages.In addition, the fluorescence probe has obvious advantages in selectivity, sensitivity and nanoscale spatial resolution.Methods: Rhodamine B was used as the basic skeleton of the probe, and the target product was synthesized by connecting the heterocyclic compounds with different structures. The fluorescence properties of the compounds were studied by fluorescence spectrophotometer.The cytotoxicity and imaging properties were also studied.Results: a fluorescent molecular probe, Amonobenzene, was successfully synthesized, and its structure was confirmed by 1HNMR-13CNMR.The fluorescence intensity of probe D decreases with increasing pH from 4.26 to 7.26 in the Britton-Robinson-BR) buffer solution, and the color of the solution gradually decreases and disappears.In the reversibility experiment, the fluorescence intensity can return to the original value when the pH value changes back and forth between 4.26 and 6.98, and the fluorescence intensity can be recycled at least six times.In the experiments of resistance to interference of metal ions, the metal ions have little effect on the recognition of probe D, regardless of whether the pH value is 7.26 or 4.26.However, when the probe D was co-cultured with PC12MDA-MB-231 cells, the low concentration of probe D caused less damage to both cells, but unfortunately, probe D did not stain the lysosomes of the cells.Conclusion: although probe D has not achieved satisfactory results in cellular imaging, its high selectivity and high sensitivity to protons can be applied to weak acid environment monitoring.
