离子色谱法测定不同产地葛根多糖中的单糖组成

【摘要】:建立了葛根中岩藻糖、鼠李糖、阿拉伯糖、半乳糖、葡萄糖、甘露糖、木糖、核糖、甘露醇、木糖醇、果糖等11种单糖的离子色谱方法,并应用此方法分析了不同产地葛根多糖中单糖组成及含量。葛根多糖经超声提取,Box-Behnken响应面优化超声提取条件,三氟乙酸水解,以Carbo Pac PA20(150 mm×3 mm)色谱柱分离,20 mmol/L NaOH溶液等度洗脱。结果表明,11种单糖的定量限为0.03~0.07 mg/kg,在0.2~5 mg/L线性范围内线性关系良好(r~20.9990),加标回收率为80.3%~112.4%。经Box-Behnken响应面优化后的最佳提取工艺为液料比28:1(mL/g),超声时间44 min,超声温度65℃。经离子色谱方法测定不同产地的葛根样品中均含有岩藻糖、鼠李糖、葡萄糖,其中葡萄糖的含量最高含量在114.3~622.2 mg/g之间。方法可为葛根质量评价提供技术支撑。
[Abstract]:A method for ion chromatography of 11 monosaccharides, such as fucose, rhamnose, arabinose, galactose, glucose, mannose, xylose, ribose, mannitol, xylitol and fructose, was established. The composition and content of monosaccharide in Pueraria lobata polysaccharides from different areas were analyzed by this method. The ultrasonic extraction conditions of Pueraria polysaccharide were optimized by ultrasonic extraction Box-Behnken response surface, trifluoroacetic acid was hydrolyzed, and iso-elution of 20 mmol/L NaOH solution was separated by Carbo Pac PA20(150 mm 脳 3 mm chromatographic column. The results showed that the quantitative limit of 11 monosaccharides was 0.03 ~ 0.07 mg / kg, the linear range was 0.2 ~ 5 mg/L and the recovery rate was 80.3% ~ 112.4%.
