二甲双胍衍生物1,3,5-三嗪类化合物的合成与机理

【摘要】:二甲双胍是胍类化合物之中十分重要的降血糖药物之一,而由其合成1,3,5-三嗪类衍生物属于用途特别广泛的含氮有机化合物,在医药、农药等领域中的地位非常关键,一直以来也是科研界所研究的热点。通过采用对二甲双胍的结构进行适当的衍生化修饰等方法,很可能会获得对细菌的抑制活性很好、并且抑菌谱广等优点的1,3,5-三嗪类衍生物,从而能够更好地应用到医学、药学、化工行业等领域。因此,本论文的研究主要包括以下三个部分:(1)利用盐酸二甲胺和双氰胺为原料来设计合成出盐酸二甲双胍,并进一步对设计方案从溶剂的选择,反应物配比的比较,反应温度对产物产率的影响,反应时间对产物产率的影响,实验处理工艺的优化等方面进行了比较系统的研究,最终确定了一种简单易行,绿色环保,特别是产品中二甲胺残留量可以控制在10ppm以下,适合进行工艺化生产的最优方案。(2)详细介绍了利用原料二甲双胍来制备1,3,5-三嗪类衍生物的合成方法,以及新制备的1,3,5-三嗪类衍生物在农药行业(除草剂)、紫外线吸收剂、医药行业,和光学领域中的用途等方面的重要的应用。合成了22个1,3,5-三嗪类衍生物,并对这三类衍生物的反应机理进行了详细的研究,采用核磁共振氢谱、红外光谱、紫外—可见吸收光谱等谱学表征。(3)通过对二甲双胍与部分过渡金属的配位进行分析,对可能的反应机理进行了相关的理论计算。探索了10个由二甲双胍衍生物与金属离子的配位反应,以及可能形成的金属配合物的基本谱学表征,探究了不同的配位金属对紫外-可见吸收光谱的吸收峰的影响。
[Abstract]:Metformin is one of the most important hypoglycemic drugs in guanidine compounds. It has always been a hot spot in the scientific research field. By means of proper derivatization and modification of metformin structure, it is very likely that the antibacterial activity of metformin will be very good, and that the derivatives of 1-triazine with wide antibacterial spectrum will be better applied to medicine and pharmacy. Chemical industry and other fields. Therefore, the research in this paper mainly includes the following three parts: 1) the metformin hydrochloride is designed and synthesized by using dimethylamine hydrochloride and dicyandiamide as raw materials, and the design scheme is further compared from the selection of solvent and the proportion of reactants. The effect of reaction temperature on the yield of the product, the effect of reaction time on the yield of the product, and the optimization of the experimental treatment process were studied systematically. In particular, the dimethylamine residue in the product can be controlled below 10ppm, and the optimum scheme for the production of dimethylamine, which is suitable for the process production, is described in detail. The synthetic method for the preparation of 1ttriazine derivatives by using metformin as raw material is introduced in detail. As well as the important applications of the newly prepared triazine derivatives in the pesticide industry (herbicide, ultraviolet absorbent, pharmaceutical industry, optical field, etc.) In this paper, 22 1-triazine derivatives have been synthesized, and their reaction mechanism has been studied in detail. Nuclear magnetic resonance spectroscopy (NMR) and infrared spectroscopy (IR) have been used to study the reaction mechanism of these three derivatives. By analyzing the coordination of metformin with some transition metals, the possible reaction mechanism was calculated theoretically. The coordination reactions of metformin derivatives with metal ions and the basic spectral characterization of metal complexes were investigated. The effects of different coordination metals on the absorption peaks of UV-Vis absorption spectra were investigated.
